N-Acetylglutamate synthase (NAGS) is an enzyme that catalyses the production of N-acetylglutamate (NAG) from glutamate and acetyl-CoA. Put simply NAGS catalyzes the following reaction: :acetyl-CoA + L-glutamate → CoA + N-acetyl- L-glutamate NAGS, a member of the N-acetyltransferase family of enzymes, is present in both prokaryotes and eukaryotes, although its role and structure differ widely depending on the species. NAG can be used in the production of ornithine and arginine, two important amino acids, or as an allosteric cofactor for carbamoyl phosphate synthase (CPS1). In mammals, NAGS is expressed primarily in the liver and small intestine, and is localized to the mitochondrial matrix.
Most prokaryotes (bacteria) and lower eukaryotes (fungus, green algae, plants, and so on) produce NAG through ornithine acetyltransferase (OAT), which is part of a ‘cyclic’ ornithine production pathway. NAGS is therefore used in a supportive role, replenishing NAG reserves as required. In some plants and bacteria, however, NAGS catalyzes the first step in a ‘linear’ arginine production pathway. The protein sequences of NAGS between prokaryotes, lower eukaryotes and higher eukaryotes have shown a remarkable lack of similarity.

Inactivity of NAGS results in N-acetylglutamate synthase deficiency, a form of hyperammonemia. In many vertebrates, N-acetylglutamate is an essential allosteric cofactor of CPS1, the enzyme that catalyzes the first step of the urea cycle. Without NAG stimulation, CPS1 cannot convert ammonia to carbamoyl phosphate, resulting in toxic ammonia accumulation. Carbamoyl glutamate has shown promise as a possible treatment for NAGS deficiency.

In contrast to prokaryotes, NAGS in mammals is enhanced by arginine, along with protamines. It is inhibited by N-acetylglutamic acid and its analogues (other N-acetylated compounds). The brain also contains N-acetylglutamic acid at trace amounts, however no expression of NAGS is found. This suggests that N-acetylglutamic acid is produced by another enzyme in the brain that is yet to be determined.

Sequence identity between prokaryotic and eukaryotic NAGS is largely <30%, while sequence identity between lower and higher eukaryotes is ~20%. Enzyme activity of NAGS is modulated by L-arginine, which acts as an inhibitor in plant and bacterial NAGS, but an effector in vertebrates. While the role of arginine as an inhibitor of NAG in ornithine and arginine synthesis is well understood, there is some controversy as to the role of NAG in the urea cycle. The currently accepted role of NAG in vertebrates is as an essential allosteric cofactor for CPS1, and therefore it acts as the primary controller of flux through the urea cycle.
The synthesis of carbamoyl phosphate and the urea cycle are dependent on the presence of N-acetylglutamic acid (NAcGlu), which allosterically activates CPS1. NAcGlu is an obligate activator of carbamoyl phosphate synthetase.Kaplan Medical USMLE Step 1 Biochemistry and Medical Genetics Lecture Notes 2010, page 261 Synthesis of NAcGlu by N-acetylglutamate synthase (NAGS) is stimulated by both Arg, allosteric stimulator of NAGS, and Glu, a product in the transamination reactions and one of NAGS's substrates, both of which are elevated when free amino acids are elevated. So Glu not only is a substrate for NAGS but also serves as an activator for the urea cycle.

A simplified reaction mechanism for N-acetylglutamate synthase (NAGS) Two mechanisms for N-acetyltransferase function have been proposed: a two-step, ping-pong mechanism involving transfer of the relevant acetyl group to an activated cysteine residue and a one-step mechanism through direct attack of the amino nitrogen on the carbonyl group. Studies conducted using NAGS derived from Neisseria gonorrhoeae suggest that NAGS proceeds through the previously described one-step mechanism. In this proposal, the carbonyl group of acetyl- CoA is attacked directly by the α-amino nitrogen of glutamate. This mechanism is supported by the activation of the carbonyl through hydrogen bond polarization, as well as the absence of a suitable cysteine within the active site to act as an intermediate acceptor of the acetyl group.

N-Acetylglutamate synthase is an enzyme that serves as a replenisher of N-acetylglutamic acid to supplement any N-acetylglutamic acid lost by the cell through mitosis or degradation. NAGS synthesizes N-acetylglutamic acid by catalyzing the addition of an acetyl group from acetyl-coenzyme A to glutamate. In prokaryotes with non-cyclic ornithine production, NAGS is the sole method of N-acetylglutamic acid synthesis and is inhibited by arginine. Acetylation of glutamate is thought to prevent glutamate from being used by proline biosynthesis.

Anglo-Scottish.border.history AD 1520s-1530s. People living along the Scottish/English border were so entangled in their dual heritage during the previous 200 years of intermittent warfare that they hardly knew or cared to which king or country they owed allegiance. Duty to family assumed utmost importance. When called upon during frequent hostilities either between the two countries or, just as often, between feuding overlords, nobility and commoner alike would don their steel helmets and pikes, saddle their "nags," as their small but robust horses were called, and head into a fray on one side or the other—but only to the extent it suited each man's own purpose.
The route splits in Manns Harbor as Bypass US 64 uses the newer and wider Virginia Dare Memorial Bridge to cross Croatan Sound, bypassing Manteo to the south. The mainline route follows the older, narrower William B. Umstead Bridge and goes through the community of Manteo before rejoining the bypass route to access a series of bridges and causeways that connect Roanoke Island to Bodie Island on the Outer Banks. US 64 terminates at Whalebone Junction, a location in Nags Head that forms the three-way confluence of US 64, US 158 and NC 12\. The traditional arms of Brochwel Ysgithrog. The arms later assigned by the College of Arms to Brochwel, and that can be used by his male heirs, are ‘Sable, three nags' heads, erased argent’ which may represent three beheaded Saxon white horses. Many later tribes and family lines in the area claim descent from Brochwel and include his arms within theirs. Most of the genealogies of these families were first documented by the heralds in the 16th century when the view taken of Brochwel can be illustrated by the following quotation: Pengwern was certainly a Welsh kingdom or Royal residence which appears to have been located somewhere in Shropshire.
It > was a hedge alehouse, where the border farmers of either country often > stopped to refresh themselves and their nags, in their way to and from the > fairs and trysts in Cumberland, and especially those who came from or went > to Scotland, through a barren and lonely district, without either road or > pathway, emphatically called the Waste of Bewcastle." Gillesland - a brief Historical and Statistical Notice of its Locality and Mineral Waters > "This is an exact description of Mumps Ha’ as it existed till the year 1831.

The ship, en route to Cuban waters for survey duty, foundered shortly after 01:00 on the 24th. He is a man of almost superhuman strength, and was renowned in the country for his work in the fields. After being taken from his village, he eventually settles into life in the city, and, while his presence inspires fear in the other servants, he is able to remain on at least cordial terms with them. During this time, Gerasim becomes infatuated with Tatiana, the mistress’ laundress. He offers her gifts, including a gingerbread chicken, and follows her, smiling and making his characteristic unintelligible noises. His affection is quite protective, and he threatens a servant who “nags” her too severely. In another incident, Kapiton Klimov, the widow's shoemaker, speaks “too attentively” with Tatiana, and is, too, threatened by Gerasim.
George MacDonald Fraser, p.38 When raiding, or riding, as it was termed, the reivers rode light on hardy nags or ponies renowned for the ability to pick their way over the boggy moss lands (see: Galloway pony, Hobelar). The original dress of a shepherd's plaid was later replaced by light armour such as brigandines or jacks of plate (a type of sleeveless doublet into which small plates of steel were stitched), and metal helmets such as burgonets or morions; hence their nickname of the "steel bonnets". They were armed with light lances and small shields, and sometimes also with longbows, or light crossbows, known as "latches", or later on in their history with one or more pistols.
Extrajudicial killings (EJKs) is also synonymous with the term "extralegal killings" (ELKs). Extrajudicial/ extralegal killings (EJKs/ ELKs) and enforced disappearances (EDs) are unique in the Philippines in as much as it is publicly and commonly known to be committed also by non-state armed groups (NAGs) such as the New Peoples Army (NPA) and the Moro Islamic Liberation Front (MILF). Although cases have been well documented with conservative estimates of EJKs/ ELKs and EDs committed by the NPAs numbering to about 900–1,000 victims based on the discovery of numerous mass grave sites all over country, legal mechanisms for accountability of non-state actors have been weak if not wholly non-existent.
When lounge singer Dean Martin (Northam) and burlesque comic Jerry Lewis (Hayes) clicked during an impromptu pairing in 1946, neither anticipated their unparalleled success as a team. Lewis was going nowhere fast with an act built around miming to records, while Martin was making a subpar living as a crooner, sleeping on his agent's couch. Lewis had beseeched his agent to reteam him with the reluctant Martin; Lewis instigated their act and adopted the jaded pro as his unofficial big brother. Ever aloof, Martin is content to cash in on the bonanza and continue his sideline in womanizing. But as Lewis assumes more control, Martin’s mistress, Jeanne (Levering), nags him to take more credit.

Carbamoyl phosphate synthase I is an enzyme found in mitochondrial matrix and it catalyzes the very first reaction of the Urea cycle, in which carbamoyl phosphate is produced. Carbamoyl Phosphate Synthase 1, abbreviated as CPS1, is activated by its natural activator N-Acetyl glutamate, which in turn is synthesized from acetyl-CoA and glutamic acid in the reaction catalyzed by N-Acetyl glutamate synthase, commonly called NAGS. N-Acetyl Glutamate is required for the Urea cycle to take place. Deficiency in N-Acetylglutamate synthase or a genetic mutation in the gene coding for the enzyme, will lead to urea cycle failure in which ammonia is not converted to urea, but rather accumulated in blood leading to the condition called Type I Hyperammonemia.
When the boat was half a mile off Nags Head (a promontory at the southern tip of the southeastern peninsula of St. Kitts), and entering the rougher seas that line up with the channel between the two islands, the ferry boat took on water and sank. In 1992, after 14 years with Southern Living he left to pursue freelance photography and to focus on contributing to various books, especially books on the lighthouses of North Carolina which became his passion. Annual round-ups on common land were enforced, and any stallion under the height limit was ordered to be destroyed, along with "all unlikely [small horses] whether mares or foals". Henry VIII also established a stud for breeding imported horses such as the Spanish Jennet, Neapolitan coursers, Irish Hobbies, Flemish "roiles", or draught horses, and Scottish "nags", or riding horses. However, it was reported in 1577 that this had "little effect"; soon after, in the reign of Queen Elizabeth I, Nicholas Arnold was said to have bred "the best horses in England". During the successive reigns of queens Mary I and Elizabeth I, laws were introduced with the aim of reducing horse theft, requiring all sale transactions of horses to be recorded.

Houses along High Street are a mix of thatched cottages and Bedfordshire brick dwellings, with an assortment of renovated or rebuilt barn buildings in keeping to some extent with earlier farm courtyard structures. Due to closures, there is now only one public house in Greenfield called The Compasses. Three former pubs, were the Swan Beerhouse on Mill Lane which closed in 1909, the Nags Head Beerhouse on the High Street which closed in 1913, and the Old Bell Public House which closed more recently in 2007.

In Wilmington, sustained winds of 25 mph (40 km/h) and gusts of 28 mph (45 km/h) were reported, as well as tides 2–3 feet (0.6-0.9 m) above normal.
Tropical Storm Beryl off the North Carolina on July 19 The depression moved slowly to the north-northwest through a break in the subtropical ridge, and as convective banding features became more prominent the system intensified into Tropical Storm Beryl. Throughout much of its duration, the storm tracked through an environment with light vertical wind shear and well-established upper-level outflow. Shortly after becoming a tropical storm, the low-level circulation of Beryl became exposed with limited deep convection, though deep convection re-developed the following morning. Outflow continued to improve, and with warm sea surface temperatures Beryl gradually intensified to attain peak winds of 60 mph (95 km/h) early on July 20 while located about 120 miles (190 km) east of Nags Head, North Carolina.

In 1950, Williams produced Lucky Inning, their first pinball machine to have its bottom flippers facing inward in the modern manner. In 1948, Williams sold 49% of his company to a Philadelphia distributor named Sam Stern, who became a vice president and took over much of the day-to-day running of the company. In 1959, Stern orchestrated a buyout of Williams by Consolidated Sun Ray, a New York retail conglomerate that operated drug stores and discount houses. Harry Williams left the company after the buyout of his company, which was renamed the Williams Electronic Manufacturing Corporation.Smith 2019, pp. 92 In 1960, Harry Williams designed his last pinball machine for Williams, the horse racing-themed Nags. The last game he designed for Williams was also one of the last electromechanical games, Rancho (1977). In 1961, Stern bought Williams back from Consolidated Sun Ray.
It has a claimed date of 1189, based on the fact it is constructed on the site of the Nottingham Castle brewhouse; the present building dates from around 1650. Likewise, The Nags Head in Burntwood, Staffordshire only dates back to the 16th century, while it has been claimed that a pub on the site is mentioned in the Domesday book, Burntwood is not in fact listed. There is archaeological evidence that parts of the foundations of The Old Ferryboat Inn in Holywell may date to AD 460, and there is evidence of ale being served as early as AD 560. The Bingley Arms, Bardsey, Yorkshire, is claimed to date to 905 AD. Ye Olde Salutation Inn in Nottingham dates from 1240, although the building served as a tannery and a private residence before becoming an inn sometime before the English Civil War.
There being no weight limits on the village lanes in this corner of Essex, this narrow bridge has become somewhat degraded over recent decades, by the passage of large trucks and of other vehicles driven with insufficient precision between its parapets. There is no modern road, at this point, following the route of the Roman road out of the village, but the old road's route can be followed at Tawney Common a few miles to the south west. Modern Moreton remains at the confluence of various minor roads, however, which link to Fyfield, to the east, Bobbingworth to the south, High Laver (burial place of John Locke) to the west and Matching to the north. Where the roads meet there remain several substantial village houses from the late Medieval / early modern period of English history, as well as two traditional English pubs, being The White Hart and The Nags Head.

A team of Allied prisoners of war (POWs), coached and led by English Captain John Colby (Michael Caine), a professional footballer for West Ham United before the war, agree to play an exhibition match against a German team, only to find themselves involved in a German propaganda stunt. Colby is the captain and essentially the manager of the team and thus chooses his squad of players. Another POW, Robert Hatch (Sylvester Stallone), an American who is serving with the Canadian Army, is not initially chosen, but eventually nags the reluctant Colby into letting him on the team as the team's trainer, as Hatch needs to be with the team to facilitate his upcoming escape attempt. Colby's superior officers repeatedly try to convince Colby to use the match as an opportunity for an escape attempt, but Colby consistently refuses, fearing that such an attempt will only result in getting his players killed.
Roger Blin, in an interview on 2 March 1975, in Paris, said: "Beckett absolutely didn't want me to try to do Embers for the theatre because, when you listen, you don't know if Ada exists or not, [or] whether she only exists in the imagination of the character Henry."Oppenheim, L., Directing Beckett (Ann Arbor, University of Michigan Press), p 310 Ada is "immensely there", though, her personality is allowed to shine throughout her conversation with Henry; she doesn’t merely respond, she initiates lines of thought, she nags him like a mother with her list of don’ts, jokes with him, reproves him in a matter-of-fact way and refuses to mollycoddle him. She doesn’t appear to take him very seriously either. Henry is obviously incapable of imagining her any other way than how she was when they were together, further evidence of his declining creative powers.
