The cross talk between MID1 and the Shh pathway is also supported by experimental evidence in model organisms.
These transcription factors are induced sequentially along the SHH concentration gradient with respect to the amount and time of exposure to SHH ligand. As each population of progenitor cells responds to the different levels of SHH protein they begin to express a unique combination of transcription factors that leads to neuronal cell fate differentiation. This SHH-induced differential gene expression creates sharp boundaries between the discrete domains of transcription factor expression which ultimately patterns the ventral neural tube. The spatial and temporal aspect of the progressive induction of genes and cell fates in the ventral neural tube is illustrated by the expression domains of two of the most well characterized transcription factors Olig2 and Nkx2.2.
SHH in particular is needed for growth of epithelial cervical loops where the outer and inner epitheliums join and form a reservoir for dental stem cells. After the primary enamel knots are apoptosed, the secondary enamel knots are formed. The secondary enamel knots secrete SHH in combination with other signaling molecules to thicken the oral ectoderm and begin patterning the complex shapes of the crown of a tooth during differentiation and mineralization. In a knockout gene model absence of SHH is indicative of holoprosencephaly.
The ZPA forms where the Hox8 region joins the AER. 250px These regions are dependent on signaling in order for the appropriate induction events to occur. The AER expresses FGF8 which induces Shh expression in the posterior mesoderm. Shh then stimulates FGF4 to be expressed in the posterior part of the AER.
The secretion of SHH by the notochord establishes the ventral pole of the dorsal- ventral axis in the developing embryo.
The concentration and time-dependent cell-fate-determining activity of SHH in the ventral neural tube makes it a prime example of a morphogen. In vertebrates SHH signaling in the ventral portion of the neural tube is most notably responsible for the induction of floor plate cells and motor neurons. SHH emanates from the notochord and ventral floor plate of the developing neural tube to create a concentration gradient that spans the dorso-ventral axis and is antagonized by an inverse Wnt gradient which specifies the dorsal spinal chord. Higher concentrations of the SHH ligand are found in the most ventral aspects of the neural tube and notochord while lower concentrations are found in the more dorsal regions of the neural tube.
This suggests that the primitive carapacial ridge functioned differently and must have gained the function of mediating the ribs and carapace development later. The Pax1 and Sonic hedgehog gene (Shh) serve as key regulators during the development of the vertebral column. Shh expression in the neural tube is essential for the maintenance of Pax1 expression in the ventral sclerotome and thus plays a key role in carapacial rib development. Genetic observations of Pax1 and Shh further provide an understanding in key gene expression that could potentially be responsible for changing turtle morphology.
Shh is expressed by the contralaterally projecting RGCs and midline glial cells. Boc, or Brother of CDO (CAM-related/downregulated by oncogenes), a co-receptor for Shh that influences Shh signaling through Ptch1, seems to mediate this repulsion, as it is only on growth cones coming from the ipsilaterally projecting RGCs. Other factors influencing ipsilateral RGC growth include the Teneurin family, which are transmembrane adhesion proteins that use homophilic interactions to control guidance, and Nogo, which is expressed by midline radial glia. The Nogo receptor is only expressed by VTc RGCs.
Over expression was particularly pronounced for miR-92 and miR20a. Similarly, these trends were only present in tumours whose expression profiles revealed an activated SHH pathway. The implication is that miR-92 and other miRNAs from mir-17-92 drive the formation of human MB by acting upon the SHH/PTCH pathway. In further studies with mice, an inhibitor of the SHH pathway (cyclopamine drug) was sufficient to reduce proliferation of tumour cells that had been infected with a retro virus encoding mir-17-92 (plus a GFP reporter).
The ZLI is also characterized by a lack of lunatic fringe (lfng), which is observable even before Shh expression is observed in the forming ZLI. This indicates that cells are fated for ZLI formation before induction by Shh (n.b. Shh]' signals from the ZLI and is characteristic of the compartment during and after formation). Lfng is expressed very early (soon after gastrulation) in the region that will become the ZLI, but shortly thereafter expression recedes through migration of lfng-expressing cells to form the lfng-free wedge characteristic of the developing ZLI.
In the central nervous system (CNS), overall cell fate mapping is typically directed by the sonic hedgehog (Shh) morphogen signaling pathway. In the spinal cord, Shh is directed by both the notochord and floor plate regions which ultimately drives the organization of neural and glial progenitor populations. The specific glial populations impacted by Shh in these two regions include oligodendrocyte precursor cells (OPCs), oligodendrocytes, NG2+ cells, microglia, and astrocytes. The floor plate (FP) region of the spinal cord individually contributes to gliogenesis, or the formation of glial cells.
Although classically considered conjoined twinning (which it resembles), diprosopus is not normally due to the fusion or incomplete separation of two embryos. It is the result of abnormal activity by the protein SHH (sonic hedgehog). (The name of this protein was inspired by the Sonic the Hedgehog video game character and is part of an idiosyncratic naming tradition in molecular biology research.) SHH and its corresponding gene have been found to play an important role in signaling craniofacial patterning during embryonic development. Among other things, SHH governs the width of facial features.
If the SHH gene is mutated, the result is cyclopia, a single eye in the center of the face (Gilbert, 2000).
This gene encodes for a new transmembrane receptor and it is proposed that this receptor is an upstream regulator of SHH.
One example of an intrinsic regulator of this process is the transcription factor Ath5. Ath5 expression in retinal progenitor cells biases their differentiation into a retinal ganglion cell fate. An example of an environmental factor is the morphogen sonic hedge hog (Shh). Shh has been shown to repress the differentiation of precursor cells into retinal ganglion cells.
Holoprosencephaly is a condition most commonly caused by a mutation of the SHH gene that causes improper separation of the left and right brain and facial dysmorphia. Many systems and structures rely heavily on proper expression of the SHH gene and subsequent sonic hedgehog protein earning it the distinction of being an essential gene to development.
The cleavage is catalysed by a protease within the C-terminal domain. During the reaction a cholesterol molecule is added to the C-terminus of SHH-N. Thus the C-terminal domain acts as an intein and a cholesterol transferase. Another hydrophobic moiety a palmitate is added to the alpha- amine of N-terminal cysteine of SHH-N.
During limb bud development, down-regulation of Tbx2 fails to inhibit Shh/FGF4 signaling; therefore, resulting in increased limb bud size and duplication of the 4th digit, polydactyly. Opposite this, when Tbx2 is over expressed or duplicated, limb buds are smaller and can have reduced digit number because of the early termination of Shh and FGF4 signaling.
This can lead to the activation of GLI transcriptional activity in the cell over-secreting Shh (autocrine signaling) or in neighboring cells (paracrine signaling). Further, Shh ligands can stimulate the production of growth factors by stromal cells present in the tumor microenvironment. These growth factors, in turn, promote the growth, survival, and proliferation of cancer cells.
Shortly after the beginning of dorsoventral patterning, Shh is expressed along the basal plate (the bottom) of the embryo, which functions in "ventralization of the neural tube, promotion of growth and proliferation, and formation of the hypothalamus". As the embryo continues to develop, Shh expression characteristic of the ZLI extends dorsally to form a wedge that eventually narrows to a strip at approximately 22 somites (the number of developed rhombomeres) or less than one day in zebrafish. Although Shh expression extends dorsally from the basal plate, the ZLI is capable of forming even without the basal plate or mesodermal tissue. Shh cooperates with dlx2 and fezl anteriorly and IRX3 and dbx1a posteriorly (genes expressed vary among different organisms), which are genes that are expressed in the prethalamus and thalamus, respectively.
But, when Shh is active in the FP region, gliogenesis is activated and glial cells begin migrating to their targeted destinations to function.
The downstream targets that are activated in response to Shh pose another challenge. Genes that are targets of Shh signaling encode factors that lead to the formation of the autopod, stylopod and zeugopod. 500px Activation of Gli zinc-finger transcription factors occurs through the Hedgehog signaling pathway. There are three Gli factors that are essential for limb development: Gli1, Gli2 and Gli3.
One example is the activation of SMO without the subsequent entry of GLI transcription factors into the nucleus. Another, better characterized pathway is the activation of GLIs independent of Shh or PTCH1 / SMO. This alternate mode of triggering the activity of GLIs is common in cancer cells. Oncogenes such as KRAS can activate the GLIs in the absence of Shh signaling.
Gli and MIM have been shown to act synergistically to induce epidermal growth and MIM + Gli1 overexpressing grafts show similar growth patterns to Shh grafts.
This was shown using zebrafish mutants that had varying amounts of BMP signaling activity. Researchers observed changes in dorsal- ventral patterning, for example zebrafish deficient in certain BMPs showed a loss of dorsal sensory neurons and an expansion of interneurons. Shh secreted from the floor plate creates a gradient along the ventral neural tube. Shh functions in a concentration-dependent manner to specify ventral neuronal fates.
The spinal cord is made from part of the neural tube during development. As the neural tube begins to develop, the notochord begins to secrete a factor known as Sonic hedgehog or SHH. As a result, the floor plate then also begins to secrete SHH, and this will induce the basal plate to develop motor neurons. Meanwhile, the overlying ectoderm secretes bone morphogenetic protein (BMP).
Not only a boundary, the ZLI is also a compartment with separate cell lineage restriction boundaries both anterior and posterior of a section of shh expression. The importance of the ZLI was once more confirmed by ectopic expression of shh in other regions of the forebrain, known as the prosencephalon during development (both the telencephalon and diencephalon), inducing a ZLI-like region that induces thalamic fate.
The branching process forms the bronchi, bronchioles, and ultimately the alveoli. The four genes mostly associated with branching morphogenesis in the lung are the intercellular signalling protein – sonic hedgehog (SHH), fibroblast growth factors FGF10 and FGFR2b, and bone morphogenetic protein BMP4. FGF10 is seen to have the most prominent role. FGF10 is a paracrine signalling molecule needed for epithelial branching, and SHH inhibits FGF10.

For example, adult hippocampal neural progenitor cells need abeyance as stem cells to differentiate later. This is regulated by Shh. Shh works through a slow protein synthesis dependence, which stimulates other cascades that work synergistically with the PI3K/AKT pathway to induce proliferation. Then, the other pathway can be turned off and the effects of the PI3K/AKT pathway become insufficient in stopping differentiation.

Retinoic acid is required ventrally along with Shh to induce Pax6 and Olig2 during differentiation of motor neurons. Three main ventral cell types are established during early neural tube development: the floor plate cells, which form at the ventral midline during the neural fold stage; as well as the more dorsally located motor neurons and interneurons. These cell types are specified by the secretion of the Shh from the notochord (located ventrally to the neural tube), and later from the floor plate cells. Shh acts as a morphogen, meaning that it acts in a concentration-dependent manner to specify cell types as it moves further from its source.
Studies have suggested that while OTX2 positively restricts the ZLI anteriorly (Shh cannot be expressed where OTX2 is not), 'IRX1 negatively restricts the ZLI posteriorly. Other genes crucial for differentiation in the brain, including Fgf genes responsible for patterning of the midbrain-hindbrain boundary (MHB), have been implicated in positioning of the ZLI. Early in development the cells at the ventral midline have only been exposed to a low concentration of SHH for a relatively short time and express the transcription factor Olig2. The expression of Olig2 rapidly expands in a dorsal direction concomitantly with the continuous dorsal extension of the SHH gradient over time. However as the morphogenetic front of SHH ligand moves and begins to grow more concentrated cells that are exposed to higher levels of the ligand respond by switching off Olig2 and turning on Nkx2.2. creating a sharp boundary between the cells expressing the transcription factor Nkx2.2 ventral to the cells expressing Olig2.
The sonic hedgehog (SHH) signaling molecule assumes various roles in patterning the central nervous system (CNS) during vertebrate development. One of the most characterized functions of SHH is its role in the induction of the floor plate and diverse ventral cell types within the neural tube. The notochord a structure derived from the axial mesoderm produces SHH which travels extracellularly to the ventral region of the neural tube and instructs those cells to form the floor plate. Another view for floor plate induction hypothesizes that some precursor cells located in the notochord are inserted into the neural plate before its formation later giving rise to the floor plate.
Specifically, Sufl1 expression leads to the switch of ventral neural progenitor cells toward an oligodendroglial fate by modulating Shh distribution and increasing signaling on apical neuroepithelial cells.
Sonic hedgehog reduces the expression of Pax6. When Shh is inhibited during development, the domain of expression for Pax6 is expanded and the eyes fail to separate causing cyclopia. Overexpression of Shh causes a loss of eye structures. Retinoic acid generated from vitamin A in the retina plays an essential role in eye development as a secreted paracrine signal which restricts invasion of perioptic mesenchyme around the optic cup.
Evidence supporting this comes from studies using BMP inhibitors which ventralize the fate of the neural plate cell for a given SHH concentration. On the other hand mutation in BMP antagonists (such as noggin) produces severe defects in ventral-most characteristics of the spinal cord followed by ectopic expression of BMP in the ventral neural tube. Interactions of SHH with Fgf and RA have yet not been studied in molecular detail.
Vitronectin is another protein that is induced by SHH; it acts as an obligate co-factor for SHH signaling in the neural tube. There are five distinct progenitor domains in the ventral neural tube viz. V3 interneuron motor neurons (MN) V2 V1 and V0 interneurons (in ventral to dorsal order). These different progenitor domains are established by "communication" between different classes of homeobox transcription factors (see Trigeminal nerve).

These placodes first become evident histologically in the open neural plate. After sonic hedgehog (SHH) signalling from the notochord induces its formation, the floor plate of the incipient neural tube also secretes SHH. After closure, the neural tube forms a basal or floor plate and a roof or alar plate in response to the combined effects of SHH and factors including BMP4 secreted by the roof plate.

In addition, the fibroblast growth factor (FGF)- and sonic hedgehog (SHH)-signaling pathways have recently been reported to be able to induce cortical folds, with a full complement of cortical layers, in mice that live to adulthood. However SHH activates downstream molecules of Gli2 & Gli3. Mutant Gli2 and Gli3 embryos have abnormal development of incisors that are arrested at early in tooth development as well as small molars.
One mouse study looking at medulloblastoma growth in mice to study the Aspm gene, an ortholog to human ASPM, suggests that Aspm expression may drive postnatal cerebellar neurogenesis. This process occurs late in embryogenesis and immediately after birth over a time span of about 2 weeks in mice and 12 months in humans, and is regulated by the expression of the Shh gene. In proliferating cerebellar granule neuron progenitors (CGNPs), Shh expression in mouse models showed four times the amount of Aspm expression than those deprived of Shh expression in- vivo. This induction of Aspm and up-regulation during cerebellar neurogenesis was also seen in real-time PCR, where its expression was relatively high at the peak of neurogenesis and much lower at the end of neurogenesis.
Wnt signaling is also involved in the axis formation of specific body parts and organ systems later in development. In vertebrates, sonic hedgehog (Shh) and Wnt morphogenetic signaling gradients establish the dorsoventral axis of the central nervous system during neural tube axial patterning. High Wnt signaling establishes the dorsal region while high Shh signaling indicates the ventral region. Wnt is involved in the DV formation of the central nervous system through its involvement in axon guidance.
There are several diseases caused by defects in promoter-enhancer interactions, which is reviewed in this paper. Beta thalassemia is a certain type of blood disorders caused by a deletion of LCR enhancer element. Holoprosencephaly is cephalic disorder caused by a mutation in the SBE2 enhancer element, which in turn weakened the production of SHH gene. PPD2 (polydactyly of a triphalangeal thumb) is caused by a mutation of ZRS enhancer, which in turn strengthened the production of SHH gene.
If the SHH gene is mutated or absent the protein Sonic Hedgehog cannot do its job properly. Sonic hedgehog contributes to cell growth cell specification and formation structuring and organization of the body plan. This protein functions as a vital morphogenic signaling molecule and plays an important role in the formation of many different structures in developing embryos. The SHH gene affects several major organ systems such as the nervous system cardiovascular system respiratory system and musculoskeletal system.
Cyclopamine has a high affinity for Smoothened – and upon binding, inhibits the signal. Even though Shh may inhibit Patched, Smoothened cannot signal in the presence of Cyclopamine and thus the pathway is interrupted.
Sonic hedgehog is turned on in the posterior region via the early expression of HoxD genes, along with the expression of Hoxb8. Shh is maintained in the posterior through a feedback loop between the ZPA and the AER. Shh cleaves the Ci/Gli3 transcriptional repressor complex to convert the transcription factor Gli3 to an activator, which activates the transcription of HoxD genes along the anterior/posterior axis. It is evident that different Hox genes are critical for proper limb development in different amphibians.

The completion of the Harman Center allows the STC to expand its season to seven plays, four at SHH (which seats 755) and three at its current location, the Lansburgh Theatre (which seats 451), in addition to the Company’s annual Free For All, formerly held outdoors at Rock Creek Park’s Carter Barron Amphitheatre, but from 2009's production of The Taming of the Shrew, has been on the main stage at the Company's SHH in downtown Washington. SHH will have multiple stage configurations making it adaptable to the needs of each performance while the Harman Center will make it possible to develop the existing STC's educational programs in order to reach even more local students. Performances and programs will be made more accessible with the availability of discounted tickets.
At the same time, Wnt-7a is expressed in the dorsal ectoderm, and provides further positive feedback to FGF-4 and Shh. Without this system, limbs and digits are either significantly reduced or missing.
The level of reduction in proliferation was towards background levels of normal MB tumours: as in those uninfected with the retro virus. Once again, an aberrant SHH/PTCH pathway was necessary for these observations.
It is now known that endogenous retinoic acid acts permissively prior to limb bud initiation to allow the budding process to begin, and that the specific morphogen, hypothesized to be Shh, is normally expressed independently of retinoic acid in the posterior region of the limb bud. By looking at signaling homologs of other organisms, the segmentation gene of Drosophila, hedgehog, served as a viable candidate. Gene oscillation in presomitic cells is largely, but not completely, cell autonomous. When Notch signaling is disrupted in zebrafish, neighboring cells no longer oscillate synchronously, indicating that Notch signaling is important for keeping neighboring populations of cells synchronous. Mutations in the TP53 gene can predispose a cell to chromothripsis. Through genome sequencing of a Sonic- Hedgehog medulloblastoma (SHH-MB) brain tumour, a significant link between TP53 mutations and chromothripsis in SHH-MBs has been found. Further studies on the association between TP53 and chromothripsis has signified a role for p53, a tumour suppressor protein, in the massive genomic rearrangement which take place which takes place in chromothripsis. Hence there is a strong association between p53 status and chromothripsis, giving an insight into why some cancers are more aggressive.
CDON mutations are thought to diminish sonic hedgehog (SHH)-pathway activity which is important in stimulating cell proliferation, differentiation, and tissue patterning at multiple points in animal development. CDON was shown to play a role in differentiation of midbrain dopaminergic neurons through the interference with of Shh signaling pathway. Mutations in CDON gene has been associated with Holoprosencephaly which is structural anomaly of the brain, in which the developing forebrain fails to correctly separate into right and left hemispheres. CDON mutations synergistically interact with prenatal alcohol exposure to increase susceptibility to Holoprosencephaly.
When Shh diffuses from the ZPA, it predominates in the posterior region of the limb bud, activating Gli3 in the posterior region, while the repressor is still active in the anterior region. She and her brother, Shin Fujiyama, created incorporated Students Helping Honduras in 2007Pacific Citizen after visiting Honduras on a service learning trip. Along with the role of Wnt genes in patterning the anteroposterior axis through gradient polarization, Wnt8b is expressed within the ZLI itself and may help guide dorsal movement of Shh expression.
Mutations in the SHH gene can cause malformation of components of these systems which can result in major problems in the developing embryo. The brain and eyes for example can be significantly impacted by mutations in this gene and cause disorders such as Microphthalmia and Holoprosencephaly. Microphthalmia is a condition that affects the eyes which results in small underdeveloped tissues in one or both eyes. This can lead to issues ranging from a coloboma to a single small eye to the absence of eyes altogether.

Besides these well known dorsalizing factors, further molecules known to be involved in dorsal ventral patterning are also present in S. kowalevskii, such as a netrin that groups with netrin gene class 1 and 2. Netrin is important in patterning of the neural system in chordates, as well as is the molecule Shh, but S. kowalevskii was only found to have one hh gene and it appears to be expressed in a region that is uncommon to where it is usually expressed in developing chordates along the ventral midline.
Having said that, two functional analogs of cyclopamine have been approved by the FDA; vismodegib in 2012, and sonidegib in 2015. Furthermore, vismodegib was the first Shh pathway drug approved for treating cancer. While cyclopamine and vismodegib do not appear very similar, the development of vismodegib revealed which aspects of cyclopamine give it functionality and used those results to make vismodegib. Motor neuron formation generally occurs in the ventral neural tube, while the dorsal tube generally forms sensory cells. The notochord secretes a protein called sonic hedgehog (SHH), a key morphogen regulating organogenesis and having a critical role in signaling the development of motor neurons.
On 9 August 2015, a Jodel light aircraft, registration G-ASXU, was forced to land at Plymouth Airport due to poor weather. The lease-holders, Sutton Harbour Holdings plc, refused to let the aircraft take off from the airport's runway, citing safety concerns, and concrete blocks were placed around the aircraft to prevent it from being moved. Gilbertson's research focuses on understanding the link between normal development and the origins of cancer, with a particular focus on children’s brain tumours. He has shown that the four different subgroups of medulloblastoma, namely WNT, SHH, Group 3, and Group 4, begin from different cells in the brain and have different mutations in their DNA. His work has also shown that a combination of stem cell mutagenesis and extrinsic factors that enhance the proliferation of these cell populations ultimately determines organ cancer risk. In 2000, Gilbertson joined the St Jude Children's Research Hospital.
When BMP4 is expressed ectopically, within transgenic mice the hair follicle outer root sheath (ORS) the proliferation of the cell matrix is inhibited. BMP4 also activates hair keratin gene expression noting that BMP4 is important in the differentiation of the hair shaft. Noggin, a known inhibitor of BMP4, is found within the matrix cells of the hair bulb. Polydactyly is a spontaneous complex phenotypic variation, developed in one generation. Surrounding structures such as the notochord, neural tube, epidermis and lateral plate mesoderm send signals for somite differentiation Notochord protein accumulates in presomitic mesoderm destined to form the next somite and then decreases as that somite is established. The notochord and the neural tube activate the protein SHH which helps the somite to form its sclerotome. The cells of the sclerotome express the protein PAX1 that induces the cartilage and bone formation. The neural tube activates the protein WNT1 that expresses PAX 2 so the somite creates the myotome and dermatome.

Removing cells from the posterior region of the limb, they transplanted them to the anterior region and noticed that extra digits formed in the anterior area and these digits were mirror images to the normal ones. This posterior mesenchyme was the ZPA, which is now known to express the protein sonic hedgehog (Shh). One hypothesis is that at high concentrations, this unknown morphogen causes mesenchyme to form on the posterior side, while low concentrations induces meshenchyme to form on the anterior end.

The zona limitans intrathalamica (ZLI) is a lineage-restriction compartment and primary developmental boundary in the vertebrate forebrain (which is analogous to the human cerebrum) that serves as a signaling center and a restrictive border between the thalamus (also known as the dorsal thalamus) and the prethalamus (ventral thalamus). Sonic hedgehog (shh) signaling from the ZLI is crucial in the development of the diencephalon, which develops into the thalamus, the pretectum, and the anterior tegmental structures. The ZLI together with the prethalamus and thalamus make up the mid-diencephalic territory (MDT). rostral extension between the prethalamus and thalamus depicts the ZLI.
During development in both vertebrates and invertebrates, cell lineage restriction boundaries and signaling centers are formed in order to ensure proper differentiation of the body. Chemical signals, like shh from the ZLI, are often released from these boundaries and compartments in a concentration gradient (the chemicals are in much higher concentrations closer to the source) and confer identity to flanking regions. Other genes differentially expressed in these flanking regions aid in ensuring proper differentiation (see Signaling). Main structures of the developing brain: the prosencephalon (forebrain) consists of the telencephalon and diencephalon, the mesencephalon is the midbrain, and the rhombencephalon is the hindbrain.
It is in this way that each of the domains of the six progenitor cell populations are thought to be successively patterned throughout the neural tube by the SHH concentration gradient. Mutual inhibition between pairs of transcription factors expressed in neighboring domains contributes to the development of sharp boundaries however in some cases inhibitory relationship has been found even between pairs of transcription factors from more distant domains. Particularly NKX2-2 expressed in the V3 domain is reported to inhibit IRX3 expressed in V2 and more dorsal domains although V3 and V2 are separated by a further domain termed MN.
Induction of the floor plate during embryogenesis of vertebrate embryos has been studied extensively in chick and zebrafish and occurs as a result of a complex signaling network among tissues, the details of which have yet to be fully refined. Currently there are several competing lines of thought. First, floor plate differentiation may be mediated by inductive signaling from the underlying notochord, an axial mesoderm derived signaling structure. This is supported experimentally in chick, in which floor plate induction, as well as associative ventral nervous tissue differentiation, is mediated by the secreted signaling molecule sonic hedgehog (Shh).
Tetrapod limb development involves many signaling molecules such as FGF, BMP, SHH and WNT. The apical ectodermal ridge is a structure found at the distal most tip which becomes a key signaling center for the developing limb. Surprisingly many of the same signaling pathways known to play a role in tetrapod limb development have been found to play a role in bat forelimb development but the timing, intensity, and spatial gene expression of some orthologous genes have changed. Since mice are also mammals, it is convenient to compare morphology and development of forelimbs between mice and bats; these comparisons may elucidate the genetic basis of adaptive bat wing development.
During development, the crossing of the optic nerves is guided primarily by cues such as netrin, slit, semaphorin and ephrin; and by morphogens such as sonic hedgehog (Shh) and Wnt. This navigation is mediated by the neuronal growth cone, a structure that responds to the cues by ligand-receptor signalling systems that activate downstream pathways inducing changes in the cytoskeleton. Retinal ganglion cell (RGC) axons leaving the eye through the optic nerve are blocked from exiting the developing pathway by Slit2 and Sema5A inhibition, expressed bordering the optic nerve pathway. Ssh expressed at the central nervous system midline inhibits crossing prior to the chiasm, where it is downregulated.
GLI1 was originally isolated from a glioma tumour and has been found to be up regulated in many tumors including muscle, brain and skin tumors such as Basal cell carcinoma (BCC). DNA copy- number alterations that contribute to increased conversion of the oncogenes Gli1–3 into transcriptional activators by the Hedgehog signaling pathway are included in a genome-wide pattern, which was found to be correlated with an astrocytoma patient’s outcome. Shh and the Gli genes are normally expressed in hair follicles, and skin tumours expressing Gli1 may arise from hair follicles. The level of Gli1 expression correlates with the tumor grade in bone and soft tissue sarcomas.
Correct function of these proteins is critical for early development, and as such mutations of the genes encoding these proteins is known to result in various congenital defects. For example, mutation of ZIC2 is known to result in holoprosencephaly due to defect in the function of the organizer region (node), which leads to a defective anterior notochord (ANC). The ANC provides a maintenance signal to the Prechordal plate (PCP), thus a defective ANC results in degradation of the PCP, which is normally responsible for sending a shh signal to the developing forebrain resulting in the formation of the two hemispheres. Holoprosencephaly is the most common structural anomaly of the human forebrain.
The Hox genes, which initially establish the anterior-posterior axis of the entire embryo, continue to participate in the dynamic regulation of limb development even after the AER and ZPA have been established. Complex communication ensues as AER-secreted FGFs and ZPA-secreted Shh initiate and regulate Hox gene expression in the developing limb bud. Though many of the finer details remain to be resolved, a number of significant connections between Hox gene expression and the impact on limb development have been discovered. The pattern of Hox gene expression can be divided up into three phases throughout limb bud development, which corresponds to three key boundaries in proximal- distal limb development.

Gremlin1 (Grem1) is known for its antagonistic interaction with bone morphogenetic proteins (BMPs) in the TGF beta signaling pathway. Grem1 inhibits predominantly BMP2 and BMP4 in limb buds and functions as part of a self- regulatory feedback signaling system, which is essential for normal limb bud development and digit formation. Inhibition of BMPs by Grem1 in limb buds allows the transcriptional up-regulation of the fibroblast growth factors (FGFs) 4 and 8 and sonic hedgehog (SHH) ligands, which are part of the signaling system that controls progression of limb bud development. Grem1 regulation of BMP4 in mice embryos is also essential for kidney and lung branching morphogenesis.

Evidence supporting the notochord as the signaling center comes from studies in which a second notochord is implanted near a neural tube in vivo leading to the formation of an ectopic floor plate within the neural tube. Sonic hedgehog is the secreted protein which mediates signaling activities of the notochord and floor plate. Studies involving ectopic expression of SHH in vitro and in vivo result in floor plate induction and differentiation of motor neuron and ventral interneurons.
This gene appears to influence the development in humans of the forebrain. It is also present in mice and xenopus frogs, which suggests a long evolutionary history, and in those organisms its expression is confined to the forebrain, optic and olfactory areas. VAX1 gene is a transcription factor that has a homeodomain located in the 100-159 amino acid position and an Ala–rich region located in 216-253 amino acid position of the gene.
