For "Love Me Girl", Wilder used loops to evoke the emotions in the final scene of the movie Trainspotting—in which a character finds a sum of money in a locker—followed by an R&B; style similar to Justin Timberlake's. The song's melody was created using an acoustic guitar and MIDI notes on computer program GarageBand, to which synthetic elements and a dancehall-type beat were then added. The seventh track on Odd Blood, "Rome", features beats taken from Moroccan and Syrian dance music that Keating's wife had on her iPod, while for "Strange Reunions", Yeasayer created a short song under three minutes by exploring unconventional time signatures in different parts in the style of fellow New York City band Dirty Projectors.
Yeasayer wanted a recording place close to Brooklyn, but removed from it. The band members aimed to streamline their songwriting processes by only doing what things were necessary in whatever parts without embellishing with long songs. The name was chosen after the band had a brainstorming session, where Keating wanted a strong title like Blood while Wilder aimed for something more offbeat like Coca-Cola or The King of Queens. Yeasayer followed the album's release with an extensive tour of Europe and North America until November 2010. The engineer also helped the band with advice about what other groups their evolving work sounded like. Yeasayer created music in the context of thinking about the music they liked, including interesting works from contemporaries. The aim was to craft pop songs unapologetically and embrace the genre as much as possible. Wilder has noted that Keating singing lead vocals gives songs an added extra as he has "a true pop voice", while he and Tuton do not.

Odd Blood is built around sonic experimentation with different genres and synthetic musical elements. Yeasayer tried to commit to certain styles for an entire song and "not jump around". Opener "The Children" is an electronically manipulated song created with the extensive use of subwoofers. The band members sang its harmonies through a fan and used pitch shifters and were determined to have the song as the first track, especially because Keating liked the idea of starting the album as "a departure point" from the last "very choral" song on All Hour Cymbals.

Keating was joined by guitarist and keyboardist Anand Wilder, bassist Ira Wolf Tuton, and engineer Steve Revitte, but not All Hour Cymbals drummer Luke Fasano. According to a Rolling Stone article, Odd Blood was inspired by a trip to New Zealand, where Yeasayer experimented with LSD. Keating has jokingly pointed out that the aim was to "sonically challenge Rihanna in the clubs", with an emphasis on bass, while Wilder has said that the band tried to make "a poppier album".

On 26 October 2009, it was announced on 4AD's website that profits raised by the compilation for the benefit of AIDS/HIV awareness and prevention amounted to £423,212 ($668,358), a sum that represents all the profits from worldwide sales for the first half of 2009. A Dark Was the Night Live concert followed at Radio City Music Hall on May 3, 2009. The concert was released as a DVD and included footage from the show, as well as exclusive rehearsal scenes, interviews and promotional performances featuring Andrew Bird, Dirty Projectors, The National, Colin Meloy, David Sitek and Yeasayer - plus more from David Byrne, Bon Iver, My Brightest Diamond, Sharon Jones & The Dap-Kings and Feist.
Interest first arose in Glass Animals after the release of their debut extended play Leaflings on independent label Kaya Kaya Records, a subsidiary and imprint of XL Recordings, part of the Beggars Group of labels. After they became the first act to be signed to Wolf Tone, the record label of British producer Paul Epworth, they released their second, self-titled EP. A song from the EP, "Black Mambo", in addition to a re-recorded version of "Cocoa Hooves" from Leaflings, appears on Zaba, expanding the promotion of the album. Glass Animals also gained exposure in Europe by playing opening act on European shows of St. Vincent, Metronomy, Yeasayer and others. Zaba performed particularly well on the Australian charts, following three headline shows in the country and an appearance and regular airplay on Australian radio station Triple J. The album reached number 12 on the ARIA Charts.
After Dystopia the band located to a beach house on the east coast of Australia to work on new music, having collected a lot of ideas while touring for the previous 2 years. After experimenting with a range of analogue gear and new digital sounds, a fresh batch of songs were written with a more 'prog' / 'layered' sound – working with a lot of ambient layers. The band then entered a range of studio's with engineer Chris Moore (TV on the radio, yeah yeah yeah's, yeasayer), and started tracking the new record . Fresh out of the studio, they were then asked to perform at a range of festivals in Buenos Aires and Australia (Splendour in the Grass) and also a tour supporting the Flaming Lips, road- testing new material including early versions of 'Vital Signs' (then called United Illuminations), and 'This New Technology' – allowing the songs to further develop.

NBC's Law & Order aired its own fictionalized version of Lacks's story in the 2010 episode "Immortal," which Slate referred to as "shockingly close to the true story" and the musical groups Jello Biafra and the Guantanamo School of Medicine and Yeasayer both released songs about Henrietta Lacks and her legacy. Members of the Lacks family authored their own stories for the first time in 2013 when Lacks's oldest son and his wife, Lawrence and Bobbette Lacks, wrote a short digital memoir called "Hela Family Stories: Lawrence and Bobbette" with first-hand accounts of their memories of Henrietta Lacks while she was alive and of their own efforts to keep the youngest children out of unsafe living environments following their mother's death. The HeLa Project, a multimedia exhibition to honor Lacks, opened in 2017 in Baltimore at the Reginald F. Lewis Museum of Maryland African American History & Culture. It included a portrait by Kadir Nelson and a poem by Saul Williams.

Like 10 seconds after the first dude got the first envelope, I think he was in Belgium or the Netherlands or something, that guy was sharing it, and there it goes." The rest of the album was leaked via an online scavenger hunt where the band gave fans clues as to where each song and an accompanying visual directed by Yoshi Sodeoka could be found. To further promote the album, Yeasayer performed single "Longevity" on Late Night with Jimmy Fallon the night before its commercial release. In much the same way that Odd Blood was a departure from All Hour Cymbals, Fragrant World saw the band continuing to expand their sound. According to Wolf Tuton, while Odd Blood featured pop influences, Fragrant World deliberately shied away from them: "If the tone of something veered too closely to a random pop song or to something on our previous records, we’d immediately steer it in a different direction. It’s easy to swerve back into your comfort zone without even realizing it, so we were very conscious of trying to stay away from that and take this into some different sonic direction.
