Each tablet featured a yellow half which contained an immediate-release formulation of pentobarbital sodium, and a blue half which contained a sustained-release formulation of methamphetamine hydrochloride (Desoxyn Gradumet). The three dosages of tablets that were manufactured included 5mg, 10mg, and 15mg of methamphetamine hydrochloride with 30mg, 60mg, and 90mg of pentobarbital sodium, respectively. Abuse of Desbutal often involved physical separation of these two halves by sawing with a razor or fingernails, followed by the consumption of each active ingredient independently rather than concurrently.

Desbutal was marketed as an antidepressant as well as a medication for the treatment of obesity, narcolepsy, parkinsonism, and alcoholism, although it was commonly also prescribed off-label for miscellaneous ailments. It was highly abused and is no longer manufactured.

Methamphetamine (contracted from ) is a potent psychostimulant of the phenethylamine and amphetamine classes that is used to treat attention deficit hyperactivity disorder (ADHD) and obesity. Methamphetamine exists as two enantiomers, dextrorotary and levorotary. Dextromethamphetamine is a stronger CNS stimulant than levomethamphetamine; however, both are addictive and produce the same toxicity symptoms at high doses. Although rarely prescribed due to the potential risks, methamphetamine hydrochloride is approved by the United States Food and Drug Administration (USFDA) under the trade name Desoxyn.

One of the earliest uses of amphetamine and methamphetamine was during World War II, when they were used by Axis and Allied forces. As early as 1919, Akira Ogata synthesized methamphetamine via reduction of ephedrine using red phosphorus and iodine. Later, the chemists Hauschild and Dobke from the German pharmaceutical company Temmler developed an easier method for converting ephedrine to methamphetamine. As a result, it was possible for Temmler to market it on a large scale as a nonprescription drug under the trade name Pervitin (methamphetamine hydrochloride).

After the 2001 elections, Barbers authored the law creating the Philippine Drug Enforcement Agency. He was instrumental in the crafting of other laws that increased penalties for "jueteng" protectors and operators and strengthened the anti-drugs campaign, such as including possession of 10 grams of methamphetamine hydrochloride as a nonbailable offense. Barbers was a strong advocate of the death penalty for capital offenders.

The company became especially known for the introduction of its methamphetamine-hydrochloride preparation brand Pervitin, which the company produced from 1938 up until 1988. According to a Der Spiegel article in 2005, Nazi Germany believed that Pervitin could also help win World War II, so the German armed forces was supplied with more than 38 million Pervitin tablets, especially during Germany's "Blitzkrieg" invasion of Poland and the Battle of France during 1939/40 where it was introduced to soldiers to attenuate anxiety and increase performance and concentration. In 1945 the facility in East Berlin was occupied and disassembled. The company had to be completely reassembled in Hamburg. In 1960 the whole company transferred from Hamburg to Marburg.

One meth legend refers to the method of administration in which the user will heat/melt crystal methamphetamine and inhale the resulting methamphetamine vapor. The legend states that the drug, once inhaled, will re- crystallize in large amounts inside the lungs, damaging them in the process. This is a false claim as crystallized methamphetamine is always in the form of a salt (usually methamphetamine hydrochloride), which is highly soluble in water, as well as hydrophilic, and is instantly absorbed into the user's bloodstream via the alveoli. The prevalence of illegal drug use in the Philippines is lower than the global average, according to the United Nations Office on Drugs and Crime (UNODC). President Rodrigo Duterte has claimed that the country could become a "narco- state". Two of the most used and valuable illegal drugs in the country are methamphetamine hydrochloride (known locally as shabu) and marijuana. In 2012, the United Nations said the Philippines had the highest rate of methamphetamine use in East Asia, and according to a U.S. State Department report, 2.1 percent of Filipinos aged 16 to 64 use the drug based on 2008 figures by the Philippines Dangerous Drugs Board.

About nine Chinese drug cartels are involved on most illegal drug trade in the Philippines. The U.S. Department of State found out that Chinese drug cartels are behind the trade of methamphetamine hydrochloride on the Philippines. The president divulged the names of the members of a large Chinese triad group in an interview with PTV-4 on July 7. The members of the triad group included Chinese drug lords, namely Wu Tuan, aka tatay Co, Jameson Ching, Peter Lim, aka tiger balm, and Herbert Colangco, with the three under the protection of Marcelo Garbo Jr, one of the Philippine National Police generals named by Duterte on July 5.

In the early dawn of December 22, 2006, and barely four months in PDEA, Marcelino led a composite group of PDEA agents and Chinese authorities from China National Narcotics Control Commission (CNNCC) in steering the whole operations from intelligence work to the actual raid of the clandestine drug laboratory in Calumpit, Bulacan whereby they seized a 1.5 billion-peso (US$30.3 million) worth of methamphetamine hydrochloride[2]. In so doing, his team then practically dismantled one of the biggest drug syndicates in Southeast Asia as cited by the CNNCC[3]. Three members of the Chinese drug syndicate were also nabbed at Ninoy Aquino International Airport (NAIA) when they were about to leave the country. CNNCC also arrested 13 other members of the syndicate in China and recovered one ton of precursors.

A Desbutal overdose typically becomes life-threatening as a result of acute toxicity from pentobarbital poisoning, since the oral median lethal dose (LD50) of pentobarbital sodium is reached significantly sooner than the oral LD50 of methamphetamine hydrochloride when consumed as dosage units with the mass ratio present in Desbutal. Barbiturate overdose is a medical emergency which was treated with immediate gastric lavage, mechanical ventilation, and intermittent intravenous bolus administration of multiple different drugs, since no selective barbiturate binding site receptor antagonists were clinically available as an antidote, and dosages were dependent on individual patient response. These drugs often included picrotoxin, amiphenazole (Daptazile), bemegride (Megimide), and methamphetamine sulfate (Methedrine) in dosages sometimes exceeding 250mg over the first 24 hour period of being admitted into an intensive care unit (ICU). While a quantitative relationship between the blood levels of barbiturate derivatives and the depression of the central nervous system (CNS) had become established by researchers in 1955, they also emphasized that the accurate correlation of these factors is complicated by the presence of tolerance to the drug, intercurrent disease and senility, as well as the concurrent administration of other noxious substances.
