In his book PiHKAL: A Chemical Love Story, Alexander Shulgin lists the dosage range as 10 to 30 mg. 2C-T-7 is generally taken orally, and produces psychedelic and entactogenic effects that last 8 to 15 hours. Up until Operation Web Tryp and three deaths, two of which involved the use of other drugs in addition to 2C-T-7, and one which involved an excessive insufflated dose, 2C-T-7 was sold commercially in Dutch and Japanese smartshops and online.

The intraperitoneal of bufotenin is between 200–300 mg/kg (in rodents) with death occurring by respiratory arrest. The LD50 in rodents scales to between 10,000 mg and 15,000 mg for a small 50 kg (110 lb) adult.

Based on the intraperitoneal LD50 for rodents, at 74 mg per gram, it would require approximately 135 grams of beans to reach the estimated LD50of bufotenin for a 50 kg (110 lb) adult. The beans have been found to contain up to only 0.04% 5-MeO-DMT and 0.16% DMT. The leaves and bark also contain small amounts of DMT, 5-MeO- DMT and related compounds.Schultes 1976 Schultes 1977 Pachter et al. 1959 At up to 0.04% (0.4 mg per gram) 5-MeO-DMT, an effective light 5 mg dose of insufflated 5-MeO-DMT would require over 12 grams of beans. It would be extremely difficult to insufflate such a quantity, as tolerance would likely develop before the 12-gram nasal intake could be completed.

Heated humidification of respiratory gases during surgery has been demonstrated to reduce the fall in core body temperature, especially in surgeries lasting longer than one hour. The lungs can be insufflated with respiratory gases that are heated to near body temperature and humidified to 90 to 100% relative humidity(RH). Normally, air in the lungs is at core body temperature and at close to 100% RH. Especially when cold dry gases (such as anhydrous compressed gas from oxygen tanks) are used, it cool and can dry the airway. The body then utilizes energy to evaporate sufficient water from the lungs to maintain lung gas temperature and humidity.

Two lateral 5(or 3)mm operative ports are then inserted under visual guidance for performance of surgery.

The cocaine typically is poured onto a flat, hard surface (such as a mirror, CD case or book) and divided into "bumps," "lines" or "rails," and then insufflated. The amount of cocaine in a line varies widely from person to person and occasion to occasion (the purity of the cocaine is also a factor), but one line is generally considered to be a single dose and is typically 35 mg (a "bump") to 100 mg (a "rail"). As tolerance builds rapidly in the short- term (hours), many lines are often snorted to produce greater effects.
