Partial-thickness tissue engineering uses one type of cell layer, this can be in monolayers or multilayers. Monolayer epithelial sheets suffice for the study of the basic biology of oral mucosa, for example its responses to stimuli such as mechanical stress, growth factor addition and radiation damage. Oral mucosa, however, is a complex multilayer structure with proliferating and differentiating cells and monolayer epithelial sheets have been shown to be fragile, difficult to handle and likely to contract without a supporting extracellular matrix.

The summer season was the busiest time for tourism and other traffic, and ran from approximately 15 May to 30 September of each year. While Bonnington was big, she was not particularly fast, and the C.P.R.'s older all-wooden sternwheeler Rossland could easily outrun her. The size of Bonnington was not always an operating advantage, as her huge superstructure made her difficult to handle in a crosswind.
Beholder (foaled May 9, 2010) is an American Thoroughbred race horse and multiple Eclipse Award winner. She was purchased as a yearling by B. Wayne Hughes of Spendthrift Farm. She had an early reputation for being difficult to handle, but trainer Richard Mandella worked extensively with the fractious young horse, gradually developing her from a temperamental front-runner into a seasoned veteran who could be placed in strategic positions. The P.V.7 first flew on 22 June 1917, powered by a 35 hp (26 kW) ungeared Gnat engine, as the geared engine was unavailable. The P.V.7 proved to be tail heavy in the air and difficult to handle on the ground, with its sesquiplane layout and high lift wings being considered unsuitable for such a small aircraft. The Gnat engine proved to be extremely unreliable, with test flights being forced to remain within gliding distance of an airfield. Even if the album weren't half as much fun as it is, that feat would be worthy of celebration in itself.
