A pan-genome is the entire gene pool for that pathogen species, and includes genes that are not shared by all strains. Pan-genomes may be open or closed depending on whether comparative analysis of multiple strains reveals no new genes (closed) or many new genes (open) compared to the core genome for that pathogen species. In the open pan-genome, genes may be further characterized as dispensable or strain specific. Dispensable genes are those found in more than one strain, but not in all strains, of a pathogen species.

The number of core genes converges to 1,647 for number of genomes n→∞. From Donati et al. The original pangenome concept was developed by Tettelin et al. when they analyzed the genomes of eight isolates of Streptococcus agalactiae which could be described as a core genome shared by all isolates, accounting for approximately 80% of any single genome, plus a dispensable genome consisting of partially shared and strain-specific genes.
W. V. O. Quine attacked both verificationism and the very notion of meaning in his famous essay, "Two Dogmas of Empiricism". In it, he suggested that meaning was nothing more than a vague and dispensable notion. Instead, he asserted, what was more interesting to study was the synonymy between signs. He also pointed out that verificationism was tied to the distinction between analytic and synthetic statements, and asserted that such a divide was defended ambiguously.
Leafcutter ants have very specific roles for taking care of the fungal garden and dumping the refuse. Waste management is a key role for each colony's longevity. New Zealand hopes to achieve being tobacco-free by 2025 and Finland by 2040.
The Black Queen Hypothesis (BQH) is a theory of reductive evolution which suggests that, in some cases, gene loss is driven by natural selection instead of genetic drift. A gene that produces a vital biological function may become dispensable for an individual organism if the community members around it performs that function in a "leaky" fashion. Therefore, the organism gains an advantage by conserving limited resources while still acquiring necessary products like metabolites.
The TMEM39B protein interacts with the SARS-CoV-2 ORF9c accessory protein, also sometimes referred to as ORF14. ORF9C is located within the nucleocapsid (N) gene, overlapping with ORF9b. Two mutations in OFC9c resulting in premature stop codons have been observed in SARS-CoV-2 isolates, suggesting that this reading frame is dispensable for viral replication.von Brunn, A., Teepe, C., Simpson, J. C., Pepperkok, R., Friedel, C. C., Zimmer, R., ... & Haas, J. (2007).
In late G1/early S phase, the pre-RC is activated for DNA unwinding by the cyclin-dependent kinases (CDKs) and DDK. This facilitates the loading of additional replication factors (e.g., Cdc45, MCM10, GINS, and DNA polymerases) and unwinding of the DNA at the origin. Once pre-RC formation is complete, Orc1-6 and Cdc6 are no longer required for MCM2-7 retention at the origin, and they are dispensable for subsequent DNA replication.
To prevent premature degradation of p27, Skp2 levels are kept low during early and mid-G1 due to the APC/CCdh1ubiquitin ligase, which mediates the ubiquitylation of Skp2. Phosphorylation of Ser64 and, to a lesser extent, Ser72 of Skp2 contributes to the stabilization of Skp2 by preventing its association with APC/CCdh1; however, Skp2 phosphorylation on these residues is dispensable for its subcellular localization and for Skp2 assembly into an active SCF ubiquitin ligase.
In yeast, where organelle membranes are closely apposed it has been proposed that ORPs work as sterol transporters, though only a few ORPs actually bind sterols and collectively yeast ORPs are dispensable for sterol transfer in vivo. They are also part of Golgi-to-plasma membrane vesicular trafficking, but their role is not clear yet. In mammalian, ORPs participate as sterol sensors. This sensors regulate the assembly of protein complexes when cholesterol levels fluctuate.

The suit (and the Batmobile) also possesses a cloaking device that can render Terry invisible to the naked eye - although it cannot, however, filter ultraviolet light. Pads on the feet can be magnetically charged, allowing the user to cling to metal surfaces even when upside-down, mimicking bats' hangings. The suit's offensive capabilities are also significant. It carries a large number of dispensable Batarangs which, when inactive, are compact enough to be invisible to the naked eye.
Glutathione peroxidase is an enzyme containing four selenium-cofactors that catalyzes the breakdown of hydrogen peroxide and organic hydroperoxides. There are at least four different glutathione peroxidase isozymes in animals. Glutathione peroxidase 1 is the most abundant and is a very efficient scavenger of hydrogen peroxide, while glutathione peroxidase 4 is most active with lipid hydroperoxides. Surprisingly, glutathione peroxidase 1 is dispensable, as mice lacking this enzyme have normal lifespans, but they are hypersensitive to induced oxidative stress.

Long, Lexington Books, 2004, p. 82.
The sense of smell is also generally considered to be an undervalued sense, particularly in Western societies. This perspective also stems somewhat from the philosopher Immanual Kant who stated that it was the "least rewarding" and "most easily dispensable sense." In Alain Corbin's book The Foul and the Fragrant, he argues that the sense of smell is inherently tied to social power, as historically certain groups of people have been rendered as 'lesser' because of lack of hygiene.
In the 70's he played too for the National Spanish television in a few series, including a Spanish-Italian coproduction on the life of Cristóbal Colón. His last film was La trastienda (1975), an artistically dispensable but sociologically significant Spanish picture, coincident with the end of Franco era in Spain. Suárez was for many years president of the Spanish Actors Union. A tiro limpio poster (1963) He died of natural causes on August 6, 1981, in Moreda, Asturias.
In the fields of molecular biology and genetics, a pan-genome (pangenome or supragenome) is the entire set of genes for all strains within a clade. The pangenome includes: the core genome containing genes present in all strains within the clade, the accessory genome containing 'dispensable' genes present in a subset of the strains, and strain-specific genes. The study of the pangenome is called pangenomics. Some species have open (or extensive) pangenomes, while others have closed pangenomes.
A 90 kDa protein is considered fairly large for a non-fibrous protein. Hsp90 is found in bacteria and all branches of eukarya, but it is apparently absent in archaea. Whereas cytoplasmic Hsp90 is essential for viability under all conditions in eukaryotes, the bacterial homologue HtpG is dispensable under non-heat stress conditions. This protein was first isolated by extracting proteins from cells stressed by heating, dehydrating or by other means, all of which caused the cell’s proteins to begin to denature.
And preclinical studies have demonstrated the CB1 receptor is required for the behavioral effects of noradrenergic based antidepressants but is dispensable for the behavioral effect of serotonergic based antidepressants. Extrapolating from the observations that positive emotional experiences boost the immune system, Roberts speculates that intensely positive emotional experiences —sometimes brought about during mystical experiences occasioned by psychedelic medicines—may boost the immune system powerfully. Research on salivary IgA supports this hypothesis, but experimental testing has not been done.Roberts, Thomas B. (2006).
NYC saw the Putnam Division as a dispensable stepchild. The line lacked a second track, electrification, commuter parking and direct service to GCT, all of which the parallel Harlem and Hudson Divisions had, resulting in declining patronage. In 1956, the New York Central asked for permission to discontinue service on the line. On May 14, 1957, the Public Service Commission allowed for a 15 percent increase in fares, but required that service be run on the Putnam Division on a limited basis.
Workers of Atta colombica at work Leafcutter ant in Costa Rica Leafcutter ants have very specific roles in taking care of the fungal garden and dumping the refuse. Waste management is a key role for each colony's longevity. The necrotrophic parasitic fungus Escovopsis threatens the ants' food source and thus is a constant danger to the ants. The waste transporters and waste-heap workers are the older, more dispensable leafcutter ants, ensuring the healthier and younger ants can work on the fungal garden.
"Seamus" is the fifth song on Pink Floyd's 1971 album Meddle. The group performs it in the style of country blues, with vocals, an acoustic slide guitar in an open D tuning, and piano. The song is named after the Border Collie (belonging to Humble Pie leader Steve Marriott) who howls throughout the 2:15 piece. Group biographer Nicholas Schaffner calls the tune "dispensable"; David Gilmour added "I guess it wasn't really as funny to everyone else [as] it was to us".
The role of durotaxis under physiological conditions remains unknown. It may serve a purpose in fine-tuning the movement response of a cell to extracellular biochemical cues, though the relative contribution of durotaxis in a physiological environment where a cell is subject to other taxes (e.g. chemotaxis) is unknown, and may in fact prove to be wholly dispensable for cell migration in vivo. The phenomenon might also have a role in several disease states that include the stiffening of tissues, as outlined below.
Genetically engineered mice lacking NMNAT1 die during early embryogenesis, indicating a critical role of this gene in organismal viability. In contrast, mice lacking NMNAT2, which is expressed predominantly in neural tissues, complete development but die shortly after birth. However, NMNAT1 is dispensable for cell viability, as homozygous deletion of this gene occurs in glioblastoma tumors and cell lines. NMNAT enzymatic activity is probably essential at the cellular level, as complete ablation of NMNAT activity in model organisms leads to cellular inviability.
By no means had Hissette been displeased with the commission, instead he had felt honoured that such renowned tropical scientists showed an interest in his work. If the colonial administration had trusted Hissette, the commission and the “Harvard African Expedition” would have been dispensable. Confronted in 1934 by a commission in the form of the Harvard African Expedition, Hissette traveled once more to the Sankuru with the Americans as the seventh member of the expedition and showed them "his" river blindness patients.
Especially according to Easterbrook predatory pricing is rare, thus it should not be a central concern. Introducing laws for predation, especially because it is rare, could lead to generating false positive errors, which would interplay with the restriction of the rule. The main point within this argument is that government intervention is dispensable, as predation is unlikely to succeed, which creates a deterrent. This effect results by selling the products/services below the costs, which causes losses but without getting more market power.
A major theme of Virtual Light is class conflict; Warbaby and the elite are placed against Rydell and Chevette. The elite see themselves as inherently superior, and view the underclass as amoral, ruthless, and dispensable. Another important element, which is common to cyberpunk, is the idea of the power structure withholding information from the general population. The information, in this case, are the plans to rebuild an entire city completely, regardless of what its inhabitants think, reminiscent of Haussmann's renovation of Paris.
The facility was rebuilt and in places modernised in the years up to 1949. For a short period the locomotive shed experienced new life as it was allocated V 100 diesel locomotives, VT 98 railbuses and ETA 150 accumulator cars. By 1966 the routine stabling of steam locomotives had ended. With the electrification of the Ries Railway (Riesbahn), Donauwörth–Aalen, and the Ingolstadt–Neuoffingen railway many of the diesel engines were dispensable, so that in 1982 the locomotive shed was closed as an independent facility.
This introduction is closed by austere chords. This section is familiar to British television viewers as the theme of the world's longest-running TV programme (1957–present), the BBC's The Sky at Night, first presented by Sir Patrick Moore (1923-2012) and presently by Chris Lintott and Maggie Aderin-Pocock. Then the character Mélisande is introduced with characteristically strong material presented by a cor anglais solo. This is succeeded by a brief intermezzo, "At the Seashore," which Sibelius regarded as dispensable in concert performances.
Differential expression of EpoR between erythroid cells. Most notably, plasma Epo concentration is regulated by nonhematopoietic EpoR expression when the peak of plasma concentrations for induced anemia in mutant and wild-type mice. The expression of EpoR in nonhematopoietic tissue is dispensable in normal mouse development, but that the sensitivity of erythroid progenitors to Epo is regulated by the expression of EpoR. Erythropoietin mutants R103-E and S100-E (though S100 in Epo doesn't exist) has been reported to be non-erythropoietin but retain the neuroprotective function.
The process of specifying propionate is not completely understood, but it is clear that it depends on an additional protein, Dps C, which may be acting as a ketosynthase or acyltransferase selective for propionyl-CoA, and possibly Dps D makes a contribution. A dedicated MAT has been found to be dispensable for polyketide production under in vitro conditions. The PKS may "borrow" the MAT from its own fatty acid synthase and this may be the primary way ACP receives its malonate group in DXR biosynthesis.
Bauer was inclined to dismiss this as the result of "petty intruge" ("kleinliche Intrige") orchestrated by Walter Ulbricht, and refused to appreciate the dangers he faced. He was certainly not inclined to see evil in The Party, which he still saw as "the only path to a meaningful life" ("der einzige Weg zu einem sinnvollen Leben "). Bauer needed the party and the party still had a use for him, but fast moving political currents were emerging to undermine him: he was becoming all too dispensable.
Curran and his co-author Gary Ruvkun discovered approximately 60 highly conserved genes that are essential for development but can significantly increase lifespan when inactivated in adulthood. Dr. Curran’s research group has established the existence of gene-diet pairs that predict survival and aging success. The function of these genes is essential on some diets but dispensable on others [1-2]. There are potentially hundreds, if not thousands of these gene-diet pairs, which when combined, may explain the variance in aging rates across individuals.
The agency also expanded its overseas bureaus and enlisted into contracts with multiple state governments and multiple union ministries. A monopoly was again re-created and most of its competitors shut down, eventually. By late 2011, ANI accounted for about 99% of the Reuters feed and in FY 2017–18, they were paid ₹ 2.54 crore for the services. Under a new management, ANI has been accused of practicing an even-aggressive journalism model focused at maximum revenue output, where journalists were easily dispensable with.
The N-terminal 214 amino acids are dispensable for relaxation of supercoiling activity in vitro and there are four nuclear localization signals and sites for interaction with other cellular proteins within the N-terminal domain. The N-terminal domain is followed by a highly conserved, 421 amino acid core domain containing all of the catalytic residues except the active site tyrosine. This is followed by a poorly conserved linker domain of 77 amino acids. Finally there is a 53 amino acid C-terminal domain.
Tyk2−/− mice displayed a lack of responsiveness to a small amount of IFN-α, but they respond normally to a high concentration of IFN-α/β. In addition, these mice respond normally to IL-6 and IL-10, suggesting that Tyk2 is dispensable for mediating for IL-6 and IL-10 signaling and does not play a major role in IFN-α signaling. Although Tyk2−/− mice are phenotypically normal, they exhibit abnormal responses to inflammatory challenges in a variety of cells isolated from Tyk2−/− mice.
In linguistics, an adjunct is an optional, or structurally dispensable, part of a sentence, clause, or phrase that, if removed or discarded, will not otherwise affect the remainder of the sentence. Example: In the sentence John helped Bill in Central Park, the phrase in Central Park is an adjunct.See Lyons (1968). A more detailed definition of the adjunct emphasizes its attribute as a modifying form, word, or phrase that depends on another form, word, or phrase, being an element of clause structure with adverbial function.
In principle, Taiwan is currently a liberal and elections based society. Hence social versatility ought to be the standard. Notably, as per an investigation of extra cash in Taiwan by the Directorate General of Budget, Accounting and Statistics, families with the most astounding dispensable salaries number 2.6 people, while families with the least discretionary cash flow number 4.7 people. With rising costs of simple commodities and privatization of the training market, economically distraught families will end up in an undeniably hard position to educate their own children.
This is already widely deployed in the German rail network (including better frequency filters for the GSM-R radio equipment). In January 2018 the project "Digitale Schiene" (digital rail) was unveiled that intended to bring about a transition plan by mid 2018. Deutsche Bahn intends to equip 80% of the rail network with GSM-R by 2030 razing any lineside signals in the process. This will bring about 20% more trains that can be operated in the country. In the process 160,000 signals and 400,000 km of interlocking cables become dispensable.
For that, it will earn the love of the paying public and will have a successful run at the box-office." Gaurav Malani of Times of India gave the film a positive feedback and said, "For a (pleasant) change, Rohit Shetty doesn't do Golmaal 'his' style. Rather he does Golmaal in its 'original' form and that's what creates a decent difference, making Bol Bachchan fairly entertaining!". Sukanya Verma of Rediff gave the film 2.5 out of 5 stars and stated, "Bol Bachchan is dispensable cinema, forgotten almost immediately after it's over.
PI4K2A is composed of a proline-rich N-terminal region and a kinase domain located C-terminally. The proline-rich N-terminal region contains physiologically important binding sites for a ubiquitin ligase Itch and clathrin adaptor complex 3, but is likely disordered and dispensable for the kinase activity. The kinase domain can be divided into N-terminal and C-terminal lobes with the ATP binding groove and putative phosphatidylinositol binding pocket in between. The C-lobe of the kinase domain contains an additional lateral hydrophobic pocket with no distinct function assigned yet.
James Gillies, portrayed by Michael Seater, is Detective William Murdoch's nemesis. He is a cold, calculating, murderous criminal mastermind who hides behind a facade of cordiality. Gillies views his nefarious schemes as high-stakes games in which he and Murdoch match wits and, for which, there must be a clear winner and a loser. Priding himself on always being one step ahead of the detective, he often employs people as dispensable pawns to aid in his machinations, not hesitating to kill them when they no longer prove useful, to ensure their silence.
Representative examples of each of the major classes of secondary metabolites Secondary in contrast to primary metabolites are dispensable and not absolutely required for survival. Furthermore, secondary metabolites typically have a narrow species distribution. Secondary metabolites have a broad range of functions. These include pheromones that act as social signaling molecules with other individuals of the same species, communication molecules that attract and activate symbiotic organisms, agents that solubilize and transport nutrients (siderophores etc.), and competitive weapons (repellants, venoms, toxins etc.) that are used against competitors, prey, and predators.
As well as directing the film, Rilla collaborated with producer Ronald Kinnoch (using the pseudonym Geoffrey Barclay) and Stirling Silliphant on the adaptation. George Sanders co-starred with Barbara Shelley. In his other film for MGM-British, Rilla directed his father, along with George Sanders and Richard Johnson, in Cairo (1963), a remake of John Huston's The Asphalt Jungle, with Tutankhamun's jewels in a Cairo museum now the target of the robbers. His novels included Greek Chorus, The Dispensable Man, The Chinese Consortium and one simply entitled Movie.
During this time, Greider, in collaboration with Ronald A. DePinho, produced the first telomerase knockout mouse, showing that although telomerase is dispensable for life, increasingly short telomeres result in various deleterious phenotypes, colloquially referred to as premature aging. In the mid-1990s, Greider was recruited by Michael D. West, founder of biotechnology company Geron (now CEO of AgeX Therapeutics) to join the company's Scientific Advisory Board and remained on the Board until 1997. Greider accepted a faculty position at The Johns Hopkins University School of Medicine in 1997.
A special case happened in 1540. Ignatius of Loyola obtained authorization for the members of the Society of Jesus to be divided into professed with solemn vows and coadjutors with dispensable simple vows.Karl Rahner, Sacramentum Mundi, article "Religious Orders" The novelty was found in the nature of these simple vows, since they constituted the Jesuit coadjutors as religious in the true and proper sense of the word, with the consequent privileges and exemption of regulars, including them being a diriment impediment to matrimony, etc.Quanto fructuosius (1-2-1583) and Ascendente Domino (5-24-1584).
Although, RIPK1 has been primarily studied in the context of TNFR signaling, RIPK1 is also activated in response to diverse stimuli. The kinase domain, while important for necroptotic (programmed necrotic) functions, appears dispensable for pro-survival roles. Kinase activity of RIPK1 is also required for RIPK1-dependent apoptosis in conditions of IAP1/2 depletion, TAK1inhibition/depletion, RIPK3 depletion or MLKL depletion. Also, proteolytic processing of RIPk1, through both caspase-dependent and -independent mechanisms, triggers lethality that is dependent on the generation of one or more specific C-terminal cleavage product(s) of RIPk1 upon stress.
In fact, while some individual focal adhesions may display stable traction forces, others exhibit tugging traction in the manner of a repeated cycle of tugging and release. The properties of focal adhesions - whether stable or tugging - are independent of their neighbors and as such, each focal adhesion acts autonomously. This tugging traction has been shown to be dispensable to other forms of cell migration, such as chemotaxis and haptotaxis, but required for durotaxis. The focal adhesion proteins (FAK/paxillin/vinculin) - and their phosphorylation-dependent interactions as well as their asymmetrical distribution within the cell (i.e.
DDX3X performs its functions in the cell nucleus and cytoplasm, exiting the nucleus via the exportin-1/CRM1 nuclear export pathway. It was initially reported that the DDX3X helicase domain was necessary for this interaction, while the canonical features of the trafficking pathway, namely the presence of a nuclear export signal (NES) on DDX3X and Ran-GTP binding to exportin-1, were dispensable. DDX3X binding to, and trafficking by, exportin-1 has since been shown not to require the DDX3X helicase domain and be explicitly NES- and Ran-GTP-dependent.
PI4KB is composed of a proline-rich N-terminal region, a central helical domain, and a kinase domain located C-terminally. The N-terminal region contains a physiologically important binding site for a Golgi adaptor protein ACBD3, but is likely disordered and dispensable for the kinase activity. The central helical domain is responsible for the interaction with a small guanosine triphosphatase Rab11. The kinase domain can be divided into N-terminal and C-terminal lobes with the ATP binding groove and putative phosphatidylinositol binding pocket in a cleft between the lobes.
Reverse transcription of the genome occurs at a later step in the replication cycle, which results in the infectious particles having DNA rather than RNA, this also leads to less integration in the host genome. The DNA found is linear and the length of the genome. The genome encodes the usual retroviral genes pol, gag, and env as well as two additional genes tas or bel-1 and bet. The role for bet is not quite clear, research has shown that it is dispensable for replication of the virus in tissue culture.
Still in his late 20s, Jibanananda was the youngest member of the faculty and therefore regarded as the most dispensable. In the literary circle of Calcutta, he also came under serial attack. One of the most serious literary critics of that time, Sajanikanta Das, began to write aggressive critiques of his poetry in the review pages of Shanibarer Chithi (the Saturday Letter) magazine. With nothing to keep him in Calcutta, Jibanananda left for the small town of Bagerhat in the far south, there to resume his teaching career at Bagerhat P. C. College.
The impressive Abri of Laugerie-Basse, named after the village, is located on the right side of the Vézère valley, about 2 kilometers upstream from Les Eyzies. It was formed at the bottom of a 45 meters high and 500 meters long scarp slope of flat-lying limestones from the Coniacian. The 15-meter-deep Abri is located 15 meters above river level. Taking advantage of the natural conditions, the houses of Laugerie-Basse were built directly into the rocks so that building a back wall and the back half of the roof was dispensable.
A review found Mootoo's pleas for help had been ignored. A few weeks later Rosalind McGregor, 28, an Amnesty intern in Geneva, killed herself in the UK.Amnesty International has toxic working culture, report finds, The Guardian, 6 Feb 2019 According to Mootoo’s former collaborator, Salvatore Saguès, “Gaëtan’s case is merely the tip of the iceberg at Amnesty. A huge amount of suffering is caused to employees. Since the days of Salil Shetty, when top management were being paid fabulous salaries, Amnesty has become a multinational where the staff are seen as dispensable.
This gene on the X chromosome is structurally similar to a related gene on the Y chromosome. It encodes a member of the krueppel C2H2-type zinc-finger protein family. The full-length protein contains an acidic transcriptional activation domain (AD), a nuclear localization sequence (NLS) and a DNA binding domain (DBD) consisting of 13 C2H2-type zinc fingers. Studies in mouse embryonic and adult hematopoietic stem cells showed that this gene was required as a transcriptional regulator for self-renewal of both stem cell types, but it was dispensable for growth and differentiation of their progeny.
The central bus station To relieve the crowded station forecourt, a bus station was built south of the station building at the end of the 1960s, which was upgraded as the zentralen Omnibusbahnhof (central bus station, ZOB) in 1982. The bay platform track 12 was demolished to add an additional through track; this platform had become dispensable, especially since the closure of passenger and freight traffic on the branch line to Legau on 28 May 1972. In the 1970s, the track plan was simplified as a result of various rationalisations. This applied particularly to connections in the freight depot.
Our struggle in EZLN is not for women in Chiapas but for all the Mexicans. The effects of Western Capitalism makes flexibility in gender and labor roles more difficult than the indigenous cultures traditional labor. “Indigenous women’s entry into the money economy has been analyzed as making their domestic and subsistence work evermore dispensable to the reproduction of the labor force and thus reducing women’s power within the family. Indigenous men have been forced by the need to help provide for the family in the globalized capitalist economic system that favors paid economic labor while depending on female subordination and unpaid subsistence labor.
On the eve of World War II, Ivan Chonkin, the most dispensable soldier, is sent to guard a disabled military plane that crash landed on a kolkhoz (collective farm). Forgotten by his command, he earns favors of a nearby kolkhoznik woman Nyura and moves in with her. Nyura's cow eats the patch of experimental tomato-potato hybrids of the local mad genius agronomist Gladyshev, and in a retaliation the latter sends an anonymous note to NKVD that Chonkin is a deserter. When NKVDists come to arrest Chonkin, he, being a Good Soldier, refuses to leave the post, and arrests the NKVDists himself.
Mathematical fictionalism was brought to fame in 1980 when Hartry Field published Science Without Numbers,Field, Hartry, Science Without Numbers, Blackwell, 1980. which rejected and in fact reversed Quine's indispensability argument. Where Quine suggested that mathematics was indispensable for our best scientific theories, and therefore should be accepted as a body of truths talking about independently existing entities, Field suggested that mathematics was dispensable, and therefore should be considered as a body of falsehoods not talking about anything real. He did this by giving a complete axiomatization of Newtonian mechanics with no reference to numbers or functions at all.
The MYD88 gene provides instructions for making a protein involved in signaling within immune cells. The MyD88 protein acts as an adapter, connecting proteins that receive signals from outside the cell to the proteins that relay signals inside the cell. The human ortholog MYD88 seems to function similarly to mice, since the immunological phenotype of human cells deficient in MYD88 is similar to cells from MyD88 deficient mice. However, available evidence suggests that MYD88 is dispensable for human resistance to common viral infections and to all but a few pyogenic bacterial infections, demonstrating a major difference between mouse and human immune responses.
He says that assertions mediated by these forms are not distinct in meaning from the corresponding direct assertions. > They are phrases we sometimes use for emphasis or for stylistic reasons, or > to indicate the position occupied by the statement in our argument. So also > we can say 'It is a fact that he was murdered' or 'That he was murdered is > contrary to fact'. In the same context and by the same token, Ramsey cites the verbal forms (3) 'It is a fact that ___' and (4) '___ is contrary to fact' as further examples of dispensable, otiose, redundant, or purely stylistic verbiage.
In January 1942, Hayes had a knee operation which delayed his appearance at spring training and consequently caused him to gain weight. This setback along with the impressive performance by his replacement, Hal Wagner, made Hayes dispensable and on June 1, 1942, he was traded to the St. Louis Browns for Bob Harris and Bob Swift. After hitting for only a .188 batting average in 1943, it appeared as if his baseball career might be at an end when Connie Mack brought him back to the Athletics in a trade for Sam Zoldak in February 1944.
The baseline security check is an organisational instrument offering a quick overview of the prevailing IT security level. With the help of interviews, the status quo of an existing IT network (as modelled by IT baseline protection) relative to the number of security measures implemented from the IT Baseline Protection Catalogs are investigated. The result is a catalog in which the implementation status "dispensable", "yes", "partly", or "no" is entered for each relevant measure. By identifying not yet, or only partially, implemented measures, improvement options for the security of the information technology in question are highlighted.
Nef (Negative Regulatory Factor) is a small 27-35 kDa myristoylated protein encoded by primate lentiviruses. These include Human Immunodeficiency Viruses (HIV-1 and HIV-2) and Simian Immunodeficiency Virus (SIV). Nef localizes primarily to the cytoplasm but also partially to the Plasma membrane (PM) and is one of many pathogen-expressed proteins, known as virulence factors, which function to manipulate the host's cellular machinery and thus allow infection, survival or replication of the pathogen. Nef stands for "Negative Factor" and although it is often considered dispensable for HIV-1 replication, in infected hosts the viral protein markedly elevates viral titers.
Of course, few if any employees (even among those who would prefer to leave) want to be fired or laid off, so the vast majority are unwilling and unlikely to call attention to the dispensable nature of their role. As such, even if an employee has very little work to do or would only expect to be given qualitative inadequate work, s/he gives the appearance of "looking busy" (e.g., ensuring that a work-related document is open on one's computer, covering one's desk with file folders, and carrying briefcases (whether empty or loaded) from work to one's home and vice versa).
The Gypsy quarter in the Ghetto after its inhabitants had been transported to the Chełmno extermination camp. In early 1944, the ultimate fate of the Łódź Ghetto was debated among the highest- ranking Nazis. The initial wave of deportations to Chełmno ended in the autumn of 1942 with over 72,000 people defined as "dispensable" already sent to their deaths. Heinrich Himmler called for the final liquidation of the ghetto. Between 23 June and 14 July 1944, the first 10 transports of about 7,000 Jews were sent by Arthur Greiser from the Radegast train station to Chełmno.
In these cases, expressed and silent alleles differ by their methylation status, and loss of DNA methylation results in loss of imprinting and re-expression of Xist in somatic cells. During embryonic development, few genes change their methylation status, at the important exception of many genes specifically expressed in the germline. DNA methylation appears absolutely required in differentiated cells, as knockout of any of the three competent DNA methyltransferase results in embryonic or post-partum lethality. By contrast, DNA methylation is dispensable in undifferentiated cell types, such as the inner cell mass of the blastocyst, primordial germ cells or embryonic stem cells.
MyPlate replaced the USDA's 2005 MyPyramid icon, criticized as too abstract and confusing MyPlate was widely received as an improvement on the previous MyPyramid icon, which had been criticized as too abstract and confusing. The 50-percent emphasis on fruits and vegetables, as well as the simplicity and understandability of the plate image, were particularly praised. Some critics said the protein section is unnecessary, given that protein is available from other food groups, and Americans on average already eat enough; however, meat would not fit in any of the other food groups. The dairy section was criticized by some as similarly dispensable.
Such mutations are associated with the much milder Becker muscular dystrophy. Mildly affected BMD patients carrying deletions that involve over two thirds of the central rod domain have been described, suggesting that this domain is largely dispensable. Dystrophin can maintain a large degree of functionality so long as the essential terminal domains are unaffected, and exon skipping only occurs within the central rod domain. Given these parameters, exon skipping can be used to restore an open reading frame by inducing a deletion of one or several exons within the central rod domain, and thus converting a DMD phenotype into a BMD phenotype.
In archaea, RNase P ribonucleoproteins consist of 4-5 protein subunits that are associated with RNA. As revealed by in vitro reconstitution experiments these protein subunits are individually dispensable for tRNA processing that is essentially mediated by the RNA component. The structures of protein subunits of archaeal RNase P have been resolved by x-ray crystallography and NMR, thus revealing new protein domains and folding fundamental for function. Using comparative genomics and improved computational methods, a radically minimized form of the RNase P RNA, dubbed "Type T", has been found in all complete genomes in the crenarchaeal phylogenetic family Thermoproteaceae, including species in the genera Pyrobaculum, Caldivirga and Vulcanisaeta.
During the dispensing process, some of the medium is also flowing in the reverse direction (back into the reservoir). The surface tension of the fluid at the nozzle exit prevents air being sucked in and fluid from exiting the nozzle when the dispensing chamber is filled up again. The principle of this process is only useful for low-viscosity fluids and this principle is not applicable with higher fluid pressures. Ink-jet systems have the following inherent properties: • Very small single-drop volumes are achievable (8 picolitres) • High dispensing frequencies can be realised (some kHz) • Low costs for mass production • Only certain low-viscosity media are dispensable (i.e.
On April 2 Gordon scored his first goal for Toronto in a 1–1 tie against his former team Chivas. Gordon had a memorable return to The Home Depot Center on June 11 when he scored two goals against The Galaxy in a 2–2 thriller which saw two goals scored in injury time of the second half. Following Toronto's designated player signing of Danny Koevermans, Gordon became a dispensable asset and was traded along with Jacob Peterson and Nana Attakora to the San Jose Earthquakes for Ryan Johnson, allocation money, and an international roster spot in July 2011. Gordon signed a new contract with San Jose in January 2012.
Attenuation involves deleting viral genes, or gene regions, to eliminate viral functions that are expendable in tumour cells, but not in normal cells, thus making the virus safer and more tumour-specific. Cancer cells and virus-infected cells have similar alterations in their cell signalling pathways, particularly those that govern progression through the cell cycle. A viral gene whose function is to alter a pathway is dispensable in cells where the pathway is defective, but not in cells where the pathway is active. The enzymes thymidine kinase and ribonucleotide reductase in cells are responsible for DNA synthesis and are only expressed in cells which are actively replicating.
Van Duin futhermore developed himself in the genre of carnavalskraker, a dispensable comical song with mass appeal often played at the caranaval parades in the Netherlands and Flanders. This resulted in Van Duin's first chart-topping single "Willempie" in 1976, which stirred controversy for Van Duin's depiction of a retarded man. Van Duin had one more chart-topping carnavalskraker in the 1970s: "'k Heb hele grote bloemkoole", which he released as his alter ego Mr De Bok. André van Duin also had success with more serious music and released two albums dedicated to a more serious image: And're André in 1977 and And're André 2 in 1978.
The argument concept is tied to the predicate concept in a way that the complement concept is not. In linguistics, an adjunct is an optional, or structurally- dispensable, part of a sentence, clause, or phrase that, when it is removed, will not affect the remainder of the sentence except to discard from it some auxiliary information. A more detailed definition of the adjunct emphasizes its attribute as a modifying form, word, or phrase that depends on another form, word, or phrase, being an element of clause structure with adverbial function. An adjunct is not an argument or a predicative expression, and an argument is not an adjunct.
HCV IRES independently binds two components of eukaryotic translation initiation machinery, the multiprotein initiation factor eIF3 and 40S small ribosomal subunit. Moreover, it binds 40S in such a manner that AUG initiator codon is positioned in the ribosomal P-site, thus no ribosomal scanning is required. Consequently scanning factors eIF1 and eIF1A are dispensable for the HCV translation, as are components of the eIF4F complex (eIF4A, eIF4E, and eIF4G) and eIF4B, which are generally required for mRNA binding and unwinding of 5'UTR. Initiator tRNA is delivered either by eIF2 or, in stress conditions when eIF2 is inactivated, by eIF2A, eIF2D, or possibly eIF5B, a homologue of prokaryotic IF2 protein.
Though a single definition has proved elusive due to the number of potential variables and their particular interpretation under given conditions, Asle Toje claims to have found recurring traits in the research literature regarding the behavioural patterns of small powers on the international stage: # The strategic behaviour of small powers is characterized by dependence. A small power recognizes that it cannot obtain security by relying solely on its own capabilities. They cannot affect the international system alone but with some concerted effort they can affect the way the system works. A small power plays a dispensable and non-decisive part in a great power's array of political and military resources.
Other molecules fulfilling the definition of oncoantigen are EGFR/HER-1, the mucin MUC1 and the idiotype of B and T cell malignancies. Further candidates are receptor tyrosine kinases and growth factors, but in most cases the induction of effective anti-tumor immune responses against such molecules remains to be demonstrated. Most tumor antigens are not oncoantigens, either because they are intracellular molecules, like cancer-testis antigen such as MAGE family members, or because they appear to be dispensable without significant alterations of tumorigenicity, like the carcinoembryonic antigen (CEA) or the prostate specific antigen (PSA). Novel strategies will be required to identify new oncoantigens amenable to human application.
" Chrissy Williams, editor of Over the Line: An Introduction to Poetry Comics, said in an interview, "I generally say 'poetry comic' by the way, not to give any priority to the poetry, but rather to avoid any confusion with 'comic poems,' which are already their own very different hilarious genre of poem. ... the genre needs to be defined fundamentally as an inextricable combination of both comics and poetry, neither of which should be dispensable. In the best poetry comics, I believe, the piece would fail utterly (or at best be horrifically diminished) if you removed either the poetry or the artwork from the composition.
But at least the mystery's a hoot." The New York Daily News David Hinckley stated the series "dodges several bedrock problems that have torpedoed other recent attempts to make engaging series TV out of mystery thrillers", adding, "The question is whether Zero Hour can sustain [the setup] for 13 weeks, because what makes a good two-hour movie doesn't always make for 10 gripping hours of television." Mike Hale of The New York Times stated the series "is entirely dispensable, its silliness matched by its comic-book solemnity". The Washington Posts Hank Stuever called the series "rancid", adding "The dialogue is stilted and almost entirely expository.
Its ruler was Francis V, an Austrian ally and one of the many great- grandsons of the eighteenth-century Austrian empress Maria Theresa. Francis had conspicuously failed to acknowledge Napoleon III as emperor of France following the latter's successful power grab at the start of the decade. Francis of Modena had inadvertently set himself up as eminently dispensable in the eyes of Napoleon. As they traveled round the streets of Plombières, Cavour and Napoleon devised a plan to provoke an appeal for protection from the Modenese citizens to the King of Piedmont-Sardinia, and a demand for annexation of their duchy to Piedmont-Sardinia.
Fascinatingly, these two cellular populations have distinct progenitors in the fetal yolk sac suggesting that they are actual distinct in very early stages of development. They further found that TGF-beta was necessary for microglial differentiation and development, but was dispensable for border-associated macrophages. This novel discovery that microglia and border-associated macrophages possess distinct developmental programs will dramatically enhance our ability to probe the biology of these cells, specifically target them in vivo, and understand their distinct roles in disease pathogenesis. In addition to running her lab and mentoring young scientists, Greter is an Advisory Editorial Board Member for the Life Science Alliance as well as an International Advisory Board Member of the International Society of Neuroimmunology.
In 1992, India signed the Chemical Weapons Convention (CWC), stating that it did not have chemical weapons and the capacity or intent to manufacture chemical weapons. By doing this India became one of the original signatories of the CWC in 1993, and ratified it on 2 September 1996. According to India's ex-Army Chief General Sunderji, a country having the capability of making nuclear weapons does not need to have chemical weapons, since the dread of chemical weapons could be created only in those countries that do not have nuclear weapons. Others suggested that the fact that India has found chemical weapons dispensable highlighted its confidence in the conventional weapons system at its command.
Instead it crawls into your brain and makes itself at home; you'll find yourself going back to it over and over without even realizing." Chris Gibbons of XXL said, "EVOL is only the latest building block on one of the most impressive runs from any rapper we've seen, and Future shows no signs of slowing down for anything." Grant Rindner of The Line of Best Fit said, "Odds are Future will drop another project or five between now and the end of the calendar year, so while EVOL is ultimately dispensable it's still a pretty good time." Shirley Ju of HipHopDX said, "As it stands, EVOL is nothing out of the ordinary from his past endeavors.
In contrast, perturbation of glycan processing (enzymatic removal/addition of carbohydrate residues to the glycan), which occurs in both the endoplasmic reticulum and Golgi apparatus, is dispensable for isolated cells (as evidence by survival with glycosides inhibitors) but can lead to human disease (congenital disorders of glycosylation) and can be lethal in animal models. It is therefore likely that the fine processing of glycans is important for endogenous functionality, such as cell trafficking, but that this is likely to have been secondary to its role in host-pathogen interactions. A famous example of this latter effect is the ABO blood group system. Glycosylation is also known to occur on nucleocytoplasmic proteins in the form of O-GlcNAc.
The review then states: > The violence which liberal critics found so offensive has survived intact. > Aldrich sets up dispensable characters with no past and no future, as Marvin > reprieves a bunch of death row prisoners, forges them into a tough fighting > unit, and leads them on a suicide mission into Nazi France. Apart from the > values of team spirit, cudgeled by Marvin into his dropout group, Aldrich > appears to be against everything: anti-military, anti-Establishment, anti- > women, anti-religion, anti-culture, anti-life. Overriding such nihilism is > the super-crudity of Aldrich's energy and his humour, sufficiently cynical > to suggest that the whole thing is a game anyway, a spectacle that demands > an audience.
This interaction between RAPL and LFA-1 is dependent on lysine residues at positions 1097 and 1099 in the juxtamembrane region of the αL-subunit cytoplasmic domain. This is a functionally significant region of the αL cytoplasmic domain as deletion of the adjacent GFFKR motif results in a constitutively active LFA-1 integrin (124, 125). While lysines 1097 and 1099 are critical for Rap1-dependent activation of LFA-1, the β2-subunit cytoplasmic domain appears to be dispensable for activation of LFA-1 by Rap1 (126). Mutation of these lysine residues to alanine impairs the ability of LFA-1 to redistribute to the leading edge induced by Rap1 activation or overexpression of RAPL.
Pan genomics is a concept introduced in 2005 by Tettelin and Medini which eventually took root in bioinformatics. Pan genome is the complete gene repertoire of a particular taxonomic group: although initially applied to closely related strains of a species, it can be applied to a larger context like genus, phylum etc. It is divided in two parts- The Core genome: Set of genes common to all the genomes under study (These are often housekeeping genes vital for survival) and The Dispensable/Flexible Genome: Set of genes not present in all but one or some genomes under study. A bioinformatics tool BPGA can be used to characterize the Pan Genome of bacterial species.
From that, 4860 variants are identified that falls in highly conserved genomic regions # As public databases such as dbSNP and 1000 Genomes Project archive previously reported variants which are often common, it is less likely that they will contain the Miller syndrome causal variants which are rare. Hence, variants found in those data sources are filtered out and 413 variants remain. # Then, genes are assessed for whether multiple variants exist in the same gene as compound heterozygotes and 23 genes are left. # Finally, ‘dispensable’ genes are removed, those which have high-frequency non-sense mutations (in greater than 1% of subjects in the 1000 Genomes Project) which are susceptible to sequencing and alignment errors in short-read sequencing platform.
Prime editing mechanism Genomic editing takes place by transfecting cells with the pegRNA and the fusion protein. Transfection is often accomplished by introducing vectors into a cell. Once internalized, the fusion protein nicks the target DNA sequence, exposing a 3’-hydroxyl group that can be used to initiate (prime) the reverse transcription of the RT template portion of the pegRNA. This results in a branched intermediate that contains two DNA flaps: a 3’ flap that contains the newly synthesized (edited) sequence, and a 5’ flap that contains the dispensable, unedited DNA sequence. The 5’ flap is then cleaved by structure-specific endonucleases or 5’ exonucleases. This process allows 3’ flap ligation, and creates a heteroduplex DNA composed of one edited strand and one unedited strand.
There is a reference in Robert Burns's 1790 poem Tam o' Shanter to the "cotillion brent-new frae France" (brand new from France). Dancing masters differed as to the exact way of doing these dances: some, recognising the affair as an English country dance, taught that the steps and jumps of these were appropriate, while others insisted upon French elegance, recommending the basic step of the gavotte or the minuet. In reality many participants simply walked through the figure and changes, seeing these as the dance and the exact steps as dispensable. On the other hand, some figures required high skill at social dancingAldrich (1991), page 16 and many performances took place at which the majority preferred to watch rather than dance.
The basement membrane of the seminiferous tubule is a modified form of extracellular matrix composed of fibronectin, collagens, and laminin. β1- integrin is expressed on the surface of SSCs and is involved in their adhesion to the laminin component of the basement membrane although other adhesion molecules are likely also implicated in the attachment of SSCs to the basement membrane. E cadherin expression on SSCs in mice, unlike in Drosophila, have been shown to be dispensable as the transplantation of cultured SSCs lacking E-cadherin are able to colonize host seminiferous tubules and undergo spermatogenesis. In addition the blood testis barrier provides architectural support and is composed of tight junction components such as occludins, claudins and zonula occludens (ZOs) which show dynamic expression during spermatogenesis.
I was deeply moved.'quoted by Andrew Malcolm, The Remedy, Akme, 1997, p.209 In 2002, Arina Patrikova, who went on to win the 2005 Newdigate prize for poetry, reassessed the book in The Oxford Student: 'Now, more than twenty years since its completion, Making Names is neither obsolete nor dispensable. The tragicomic legal struggle does little to lessen the intellectual merits of Making Names - its prose still shines, its questions still stand, and its 'Electra' remains one of the most powerful statements of the human condition written in the last century....An attentive reader would indeed find that Making Names is easily a good novel, and it is obvious to all that it is nothing short of a film script.
Nonetheless, for the most part, their unified image served well for the Music Machine's national recognition, especially as the group made numerous appearances on the television programs Where the Action Is, American Bandstand, and Shindig!. After their long national tour, the Music Machine returned to the studio to record their debut album, (Turn On) The Music Machine. Much to the disapproval of Bonniwell, his original material had to compete with dispensable cover versions of "Cherry, Cherry", "Taxman", "See See Rider", and "96 Tears", all chosen by their record label with an expectation that the well-known songs would increase record sales. One interpretation voluntarily selected by the band was a slow, moody, fuzz-laden arrangement of "Hey Joe" which bears a strong resemblance to Jimi Hendrix's later version.
ERRα regulates genes involved in mitochondrial biogenesis, gluconeogenesis, oxidative phosphorylation, and fatty acid metabolism, and brown adipose tissue thermogenesis. It was recently identified as an important regulator of the mammalian circadian clock, and its output pathways at both transcriptional and physiological levels regulated the expression of transcription factors involved in metabolic homeostasis. It has been demonstrated that ERRα is required for the maintenance of diurnal cholesterol, glucose, insulin, bile acid, and trygliceride levels as well as locomotor rhythms in mice. ERRα is related to mitochondrial function but studies involving ERRα knockout mice suggested that this receptor, while dispensable for basal cellular function, is definitely necessary to provide the levels of energy necessary to respond to physiological and pathological insults in diverse tissues, the lack of that nuclear receptor leading to impaired fat metabolism and absorption.
As McDougall notes, he has "sought to find new formal devices within the general category of 'free verse'…Conventional but dispensable grammatical forms and punctuation disappear between intensely compressed images; subject, tense, and number are elusive; transitions are unclear; order and logic are supplied by the reader". By using such an experimental approach, Bei Dao has achieved what the poet and critic Michael Palmer has called "a poetry of complex enfoldings and crossings, of sudden juxtapositions and fractures, of pattern in a dance with randomness". This approach to poetic form earned Bei Dao his moniker as a "misty" poet, which was originally leveled pejoratively by Chinese critics who disliked his work for its lack of clarity. Some critics in the West have similarly found his work to be incomprehensible.
The feed-forward loop in the arabinose utilization systems of E.coli delays the activation of arabinose catabolism operon and transporters, potentially avoiding unnecessary metabolic transition due to temporary fluctuations in upstream signaling pathways. Similarly in the Wnt signaling pathway of Xenopus, the feed-forward loop acts as a fold-change detector that responses to the fold change, rather than the absolute change, in the level of β-catenin, potentially increasing the resistance to fluctuations in β-catenin levels. Following the convergent evolution hypothesis, the enrichment of feed-forward loops would be an adaptation for fast response and noise resistance. A recent research found that yeast grown in an environment of constant glucose developed mutations in glucose signaling pathways and growth regulation pathway, suggesting regulatory components responding to environmental changes are dispensable under constant environment.
Posner was mentioned in 2005 as a potential nominee to replace Sandra Day O'Connor because of his prominence as a scholar and an appellate judge. Robert S. Boynton wrote in The Washington Post that he believed Posner would never sit on the Supreme Court because despite his "obvious brilliance," he would be criticized for his occasionally "outrageous conclusions," such as his contention "that the rule of law is an accidental and dispensable element of legal ideology," his argument that buying and selling children on the free market would lead to better outcomes than the present situation, government-regulated adoption, and his support for the legalization of marijuana and LSD.Boynton, Robert S. Boynton. "'Sounding Off,' a review of Richard Posner's Public Intellectuals", The Washington Post Book World, January 20, 2002.
While Zhang's group was optimizing the Cas9 system in human cells, the collaborating groups of Emmanuelle Charpentier and Jennifer Doudna described a chimeric RNA design which is capable of facilitating cleavage of free-floating DNA using purified Cas9 protein and a synthetic guide. Zhang's group compared their RNA expression approach with a design based on the Doudna / Charpentier chimeric RNA for use in human cells and established features of the guide necessary for Cas9 to function effectively in mammalian cells which are dispensable in biochemical assays. His lab has developed a sensitive diagnostic nucleic acid detection protocol that is based on CRISPR termed SHERLOCK (Specific High sensitivity Enzymatic Reporter UnLOCKing) that is able to detect and distinguish strains of viruses and bacteria present in as low as attomolar (10−18 M) concentration.
In rats, G2A, similar to other pH sensing GPCRs, is located in dorsal root ganglia neurons, small diameter neurons responsible for nociception, and other nerve tissues responsible for sensing pain; it is suggested that G2A in these nerve tissues detects the acid changes that occur in the extracellular media of injured tissues and signal for the perception of pain> However, the activity of the human G2A receptor and its mouse homolog are significantly less sensitive to pH fluctuations than other pH sensing GPCRs; indeed, in studies of thymocytes and splenocytes taken from mice deficient in either the G2A or another pH- sensing GPCR, TDAG8, TDAG8 was found critical while G2A was found dispensable for sensing pH changes. Thus, the cited functions of G2A presumed due to its pH sensing ability could reflect other means for this receptor's activation.
Work on seryl-tRNA synthetase (SerRS) demonstrated for the first time that an appended domain dispensable for enzymatic activity of SerRS can be essential for the development of vertebrates. Demonstration of these broader physiological roles embedded in the genes of human aminoacyl tRNA synthetases has caused a paradigm shift in the field of human genetics and has led to new avenues for therapeutic approaches to treat a wide variety of human diseases such as inflammatory disease, autoimmune disease, cancer and neurological disorders. In 2005, Schimmel and Yang founded aTyr Pharma, a biotechnology company pursuing tRNA synthetase derived molecules, for therapeutic applications. The Yang lab has unveiled broad regulatory roles of tRNA synthetases in vascular biology, the circulatory system, stress response, and proteostasis and provided in-depth mechanistic understanding of these non- canonical functions at the levels of whole organisms, cells, and molecules.
In a more critical review, Greg Kot of the Chicago Tribune described the album as "mild", noting that Koenig's lyrical expressions of discomfort were not conveyed through the pleasant music. For Pitchfork, Mike Powell opined that the album was somewhat overlong and dispensable compared to past releases, and that its discussion of contentment and belonging felt unsuited to the group, but praised the album's braveness and new musical direction. Steven Edelstone of Paste criticized the album's lyrics, and Koenig's adoption of a "derivative" musical style that did not fit the band, concluding that "it’s simply impossible not to wonder what happened and where they lost their way, culminating in a major disappointment for perhaps the most anticipated indie rock album in recent memory". Spin's Jordan Sargent argued that the album was likely the band's worst, yet still rewarding.
After its induction, the MDO starts to orchestrate the development of the thalamic anlage by release of signalling molecules such as SHH. In mice, the function of signaling at the MDO has not been addressed directly due to a complete absence of the diencephalon in SHH mutants. Studies in chicks have shown that SHH is both necessary and sufficient for thalamic gene induction. In zebrafish, it was shown that the expression of two SHH genes, SHH-a and SHH-b (formerly described as twhh) mark the MDO territory, and that SHH signaling is sufficient for the molecular differentiation of both the prethalamus and the thalamus but is not required for their maintenance and SHH signaling from the MDO/alar plate is sufficient for the maturation of prethalamic and thalamic territory while ventral Shh signals are dispensable.
At the same time, the Research and Innovation area of the company is developing innovative hybrid technologies, which integrate getter materials into polymer matrices, initially concentrating on the development of dispensable absorbers for organic electronics applications, in particular OLED (Organic Light Emitting Diodes) light displays and sources. From 2013 onwards, further developments of the polymer technological platform have permitted the group to execute new functional polymer compounds, with the properties of interacting with the gases and optical, mechanical and surface modifying functions, according to the requirements and applications of interest, including implantable medical devices, food packaging and the field of energy storage (lithium batteries and super condensers). In 2010, it also established the company ETC, the fruit of a collaboration between CNR and SAES Getters (the majority shareholder). An innovative research programme was launched within ETC for the development of OLET technology (Organic Light Emitting Transistor).
So that, ribophorin I can keep those possible substrates in the proximity of the catalytic subunit of the enzyme; this way, the efficiency of the N-glycosylation reaction will improve during their biogenesis in the ER. But ribophorin I only changes drastically the N-glycosylation of determined substrates, as it is apparently dispensable in the same process with other substrates. When ribophorin is not essential, these precursors have shortcut to the catalytic center of the oligosaccharyltransferase or their presence depends on the rest of the non- catalytic subunits in the complex. It shows the specificity of ribophorin I for determined substrates; so that, this protein will regulate selectively the delivery of substrates to the catalytic center of the oligosaccharyltransferase complex. Although ribophorin II is still a protein quite unknown, it has been discovered that this ribophorin is part of an N-oligosaccharyltransferase complex that links high mannose oligosaccharides to asparagine residues found in the Asn-X-Ser/Thr consensus motif of nascent polypeptide chains.
In 1944, he didn't even have the right to teach anymore, was considered a "completely dispensable" teacher, and was ordered up the Rhine to build fortifications, then drafted into the Volkssturm national militia, "the oldest member of the faculty to be called up". In 1945 Heidegger wrote of his term as rector, giving the writing to his son Hermann; it was published in 1983: > The rectorate was an attempt to see something in the movement that had come > to power, beyond all its failings and crudeness, that was much more far- > reaching and that could perhaps one day bring a concentration on the > Germans' Western historical essence. It will in no way be denied that at the > time I believed in such possibilities and for that reason renounced the > actual vocation of thinking in favor of being effective in an official > capacity. Base Substitutions in the capitalized nucleotides led to the greatest reduction in FLP-mediated site-specific recombination (Wildtype x mutant and mutant x mutant): ::5'3' Many available constructs include an additional arm sequences (5'--3') one base pair away from the upstream element and in the same orientation: ::5'3' This segment is dispensable for excision but essential for integration, including Recombinase-mediated cassette exchange. Because the recombination activity can be targeted to a selected organ, or a low level of recombination activity can be used to consistently alter the DNA of only a subset of cells, Flp-FRT can be used to construct genetic mosaics in multicellular organisms. Using this technology, the loss or alteration of a gene can be studied in a given target organ of interest, even in cases where experimental animals would not survive the loss of this gene in other organs (spatial control). The effect of altering a gene can also be studied over time, by using an inducible promoter to trigger the recombination activity late in development (temporal control) - this prevents the alteration.
Xin Yi feels deeply burdened by guilt for her mistake as she struggles to stand up for herself. Despite no longer wanting to be a dispensable sticky note in other people's lives, she resolves to not fall for Cun Xi. Xin Yi's simple and selfless nature soon endears her to Cun Xi's family and, eventually, to Cun Xi. Slowly, Cun Xi begins to realize his love for Xin Yi and the child she is carrying, leaving him to wonder what life would be like if they lived together as a family. He desires to protect Xin Yi from those who would mistreat her and decides that he doesn't want her to leave him after the baby is born. Unfortunately, his epiphanies are interrupted by the unexpected return of the injured Anna. In order to conceal his marriage and impending fatherhood, Cun Xi requests that Xin Yi return to her mother’s home, until such time that he can explain the situation to Anna and his grandmother. Unfortunate events lead the devastated Anna to discover the truth while engaged in an ugly confrontation with Cun Xi’s incensed grandmother.
It held that an exemption from laws that conflicted with this was therefore mandated: > It is true that the consumption of imported meat makes such renunciation [of > meat-eating] dispensable; however, due to the fact that in this case, > personal contact with the butcher and the confidence that goes with such > contact do not exist, the consumption of imported meat is fraught with the > insecurity whether the meat really complies with the commandments of > Islam....Under these circumstances, an exemption from the mandatory stunning > of warm-blooded animals before their blood is drained cannot be precluded if > the intention connected with this exemption is to facilitate, on the one > hand, the practice of a profession with a religious character, which is > protected by fundamental rights, and, on the other hand, the observation of > religious dietary laws by the customers of the person practicing the > occupation in question. III, p.5-6 Humboldt envisioned the university education as a student-centered activity of research: > Just as primary instruction makes the teacher possible, so he renders > himself dispensable through schooling at the secondary level. The university > teacher is thus no longer a teacher and the student is no longer a pupil. > Instead the student conducts research on his own behalf and the professor > supervises his research and supports him in it.Quoted in Christopher Clark, > Iron Kingdom: The Rise and Downfall of Prussia, 1600-1947, p. 333 Early 19th-century American educators were also fascinated by German educational trends. The Prussian approach was used for example in the Michigan Constitution of 1835, which fully embraced the overall Prussian system by introducing a range of primary schools, secondary schools, and the University of Michigan itself, all administered by the state and supported with tax-based funding. However, some of the concepts in the Prussian reforms of primordial education, Bildung and its close interaction of education, society and nation- building are in conflict with American state-sceptical libertarian thinking.Wilhelm von Humboldt, Franz-Michael Konrad UTB, 21.07.2010 The main Entrance to Old College at Aberystwyth University.
